抑制血管平滑肌细胞(VSMCs)增殖既是防治血管狭窄的关键,又是延缓动脉粥样硬化(AS)性心脑血管疾病的重要举措。痰浊、血瘀是血管增生性心脑血管疾病的重要病机之一,基于中医化痰活血理论和药物拼合原理,我们把调脂(即化痰)的烟酸与活血的姜黄素进行拼合,得到了具有自主知识产权的新型化合物姜黄素烟酸酯,以期达到协同增效目的。信号通路STAT3/PTEN与VSMCs增殖密切相关,本课题组长期研究的Daxx蛋白也调节VSMCs增殖,但是否通过该信号通路还不清楚。本项目在前期研究的基础上,预采用离体细胞和整体球囊损伤大鼠模型,深入探讨Daxx/STAT3/PTEN这条新的信号通路与血管增殖的关系,以及姜黄素烟酸酯抑制VSMCs增殖的作用及机制。该项目的开展将为中西医结合防治心脑血管疾病提供一个全新的整合现代科学技术和传统中医理论的研究思路,也为血管狭窄性疾病的新药研发以及药物新靶点的揭示提供理论依据。

Inhibition of vascular smooth muscle cell (VSMCs) proliferation is a key to prevent and cure vascular stenosis, and also is very important to relief the atherosclerotic vascular disease. The phlegm-turbid and blood stasis are the important pathogenesis of vascular proliferative cardiovascular and cerebrovascular diseases. Base on Chinese huatanhuoxue theory and combination principles, we have combined nicotinic acid which can huatan and curcumin which can huoxue, and got the independent intellectual property rights of curcumin trinicotinate (CurTn). We hope CurTn is synergism to inhibite the proliferation of VSMCs. STAT3/PTEN signaling pathway is closely related to the proliferation of VSMCs, which is regulated by Daxx long-term researching in our laboratory. But whether the signaling pathway of Daxx/STAT3/PTEN mediates VSMCs proliferation is not clear. On the basis of our previous research, The project will investigate the relationship between the new signal pathway of Daxx/STAT3/PTEN and vascular proliferation by cells in vitro and balloon injury model of rats. At the same time, the project will investigate the effect and mechanism of CurTn inhibiting VSMCs proliferation. Our project will provide a new research ideas about integration of modern science and technology and traditional Chinese medicine theory for the combination of traditional Chinese and Western medicine to prevent and cure cardiovascular and cerebrovascular diseases, and new theoretical and experimental basis for prevention and treatment of vascular stenosis disease.
