Emerging infectious diseases, including Zika virus, Ebola virus and influenza virus, have raised great public health concern worldwide. The applicant combined the structural biology, biochemistry and the cellular biology methods to study the mechanisms of virus entry, and mainly focused on the influenza virus and Ebola virus. These research activities enabled the applicant to publish, thus far, 39 peer-reviewed papers in international journals. Of these publications, he was the first or corresponding author in 10 papers, including two publications in Science, one publication in Cell and Nature Reviews Microbiology, respectively. He also has 2 grants from the National Natural Science Foundation of China and the China Ministry of Science and Technology National 973 project, respectively. Virus infection includes three key steps, entry, replication and release. In the current proposal, he plan to further extend his study from virus “Surface” entry mechanism to “Internal” replication mechanism. The structures and functions of polymerases and other replication-associated proteins from Zika virus, Ebola virus and influenza virus will be systematically studied, clearly demonstrating the replication process. The goal is to gain a comprehensive view on the interspecies transmission mechanisms and the replicative features of emerging infectious viruses, which would be valuable in counteracting the virus infections and providing guidance for therapeutics development.
