申请人从事中药药理学(抗炎免疫和心血管)研究工作18年,积累了一定的创新性研究,发表SCI论文61篇,总影响因子179,H指数24,他引848次,近5年共发表SCI论文35篇,近10年在分子靶点、药理学、替代医学领域的高水平学术期刊(Oncotarget,JPET,Shock,J Ethnopharmacol,Mitochondrion等)以通讯作者名义发表了一系学术论文。近期的原创性工作主要包括:发现NF-κB信号通路关键激酶IKK-β的药物結合新靶位,基于细胞内信号分子转导通路阐释中药及其成分抗炎活性的分子机理,系统阐明人参抗心肌缺血新机制,阐释中药复方配伍减毒和中药不良反应机理。本项目将要进行的原创性工作是研究中药活性成分青藤碱治疗类风湿性关节炎的新机制,主要针对近年来新发现的炎症过程关键酶mPGES-1的调控机理,这将有助于青藤碱的临床准确定位,并拓展其临床新用途。

The applicant has been actively involved in the research of pharmacology of Chinese medicine in immune and cardiovascular systems for 18 years and obtained many innovative achievements, including 61 articles published in SCI journals (35 SCI articles in the recent 5 years) with a total impact factor of 179, 848 non-self citations, and H index of 24. Series of articles were published by the applicant as the corresponding author in the top rank SCI journals in the fields of molecular target, pharmacology, and alternative medicine, such as Oncotarget, JPET, Shock, J Ethnopharmacol, and Mitochondrion in the recent 10 years. The recent original research work of the applicant include the discovery of new drug binding sites on IKK-β in NF-κB signaling pathway, elucidation of molecular mechanisms underlying the anti-inflammatory activities of Chinese medicine and bioactive compounds isolated from Chinese medicine, illustration of new molecular mechanisms underlying the cardioprotective effects of ginseng, and the explanation of the mechanisms behind the detoxification effect of Chinese medicine by Fufang combinational use as well as behind adverse drug reaction of Chinese medicine. In this application, the innovative research on the novel molecular mechanisms underlying the anti-arthritic effects of sinomenine, an effective compound applied in the clinics for treating rheumatoid arthritis, is proposed. The research mainly focuses on the regulation of mPGES-1 by sinomenine, a newly identified key enzyme in inflammation process and an ideal drug target. This research will benefit a well-defined clinical application of sinomenine and possible extend the application of sinomenine into many other diseases.
