循环肿瘤细胞(CTCs)在判断肿瘤患者预后、评估抗肿瘤药物疗效、实时监测肿瘤基因状态和探讨肿瘤内部异质性等多个方面具有重要研究价值,是实现“实时液体活检”的有效手段之一。强生公司的CELLSEARCHTM系统是目前唯一经美国FDA批准用于临床的CTCs检测技术,除此以外,以进一步提升CTCs富集效能为目标的方法学研究层出不穷。与现有的诸多抽取血样进行富集的方法不同,血管内留置功能导丝捕捉CTCs具有可接触大量循环血液的优势。本项目基于血管内捕捉CTCs这一思路,以前期研究为基础,拟构建一组紫外光固化反应体系以实现羧酸甜菜碱甲基丙烯酸酯聚合物对介入用尼龙导丝表面的改性。在完善改性后表征、生物相容性评价及功能化实验后,利用人胃癌细胞SGC7901裸鼠原位移植瘤模型进行上述新型功能导丝富集CTCs的尝试,并与CELLSEARCHTM系统的富集结果行横向对比,期待能够实现CTCs富集方法新的突破。

Circulating tumor cells (CTCs) has its significant value in certain areas, such as predicting prognosis, evaluating therapeutic effect, monitoring gene status, and discussing intratumoral heterogeneity. CTCs is also a key to the realization of ’Real-time liquid biopsy’. The Johnson & johnson’s production CELLSEARCH system is currently the only FDA approved CTCs detection technology for clinical use. Besides, varied studies focusing on promoting the efficiency of CTCs enrichment are emerging continuously. Unlike the current CTCs detection methods based on a certain volume of blood samples, intravanous CTCs capture has its unique advantage that much larger volume of blood could be screened. This project intend to construct a UV-curing reaction system for the poly-CBMA modification of nylon wire. After the chemical characterization, biocompatibility evaluation and functional testing, CTCs capturing attempt using the newly made functional nylon wire will be conducted in nude mouse orthotopic tumor model of human gastric cancer cell line SGC7901. The comparision to CELLSEARCH system will also be involved. We hope that this project could bring us a new breakthrough in the methodology of CTCs detection .
