褪黑素是一种对人体和动植物都有重要生理功能的小分子活性物质,酿酒葡萄及葡萄酒中含有丰富的天然褪黑素,其含量已经成为衡量酿酒葡萄及葡萄酒品质的重要指标。本项目利用成熟阶段葡萄果实,通过逆境胁迫(紫外和低温)处理,测定褪黑素在逆境条件下的变化规律及其合成通路关键酶基因转录及蛋白表达差异,揭示葡萄果实成熟过程中褪黑素形成的分子机制。同时,通过逆境胁迫和外源褪黑素处理,测定葡萄多酚合成通路关键酶基因转录及蛋白表达差异,来研究褪黑素作为信号分子影响多酚合成代谢的生理机制。最后研究在果实发酵过程中,葡萄品种与酿酒酵母的互作对褪黑素的影响,从而揭示葡萄酒发酵过程中褪黑素的变化机制。最终阐明酿酒葡萄从果实成熟到发酵结束全过程中褪黑素的变化机制和影响因素,为高产褪黑素的酿酒葡萄和葡萄酒的实际生产提供一定的理论依据。

Melatonin is a widely existed small molecule bioactive component in human animals and plants which have important physiological functions. Currently research found that grape and wine are rich in melatonin and the content of melatonin has been an important paramter for measuring the quality of wine and grape. Wine grape fruits of maturity period will be used to study the forming mechanism and influence factors of melatonin under environment stress (UV and cold) in this project. The gene transcription and protein expression level of the key enzyme (TDC, T5H, AANAT, ASMT) in melatonin synthesis pathway and key enzyme in polyphenol synthesis metabolic pathway will be detected under environment stress, which aim to clarify molecular mechanism of melatonin change. In order to research the role of melatonin as signal molecule effect polyphenols biosynthesis, the key enzyme in polyphenol synthesis metabolic pathway also will be detected under environment stress and exogenous melatonin in gene transcription and protein expression level. At the same time, the influence of interaction between grape variety and Saccharomyces cerevisiae on melatonin change would be studied, which aim to explain the mechanism and influence factors of melatonin in fermentation period. It is expected that from grape ripen to wine fermentation, we can elaborate the forming mechanism of melatonin and its key influence factors, which can be a guide to improve melatonin through viticulture and wine making.
