本研究以针刺诱发神经电信号为关注点,分析针刺过程中穴位组织间隙结构和关联的化学物质的协同作用,通过动物实验设计、信号分析与数学建模相结合的方法,阐明针刺穴位信息启动和传递的路径图:(1)用胶原酶、肥大细胞稳定剂、神经阻滞剂等药物对穴位处的特定结构进行干预,检测针刺诱发外周神经电信号和化学物质(腺苷和组胺)浓度的变化,阐明穴位组织结构参与针刺(手针及电针)效应的启动和针刺关联化学物质的来源;(2)检测穴位注射腺苷、组胺诱发的外周神经电信号,并比较它们与针刺诱发神经信号的异同,阐明化学物质的针刺效应调节机制;(3)检测针刺诱发脊髓背根神经束电信号,分析针刺信息的传入机制;(4)建立描述针刺信息启动和传递机制的数学模型,对针刺穴位组织的响应进行定量分析。这些工作有助于从系统生物学角度揭示针刺的调节机制,为中医针刺理论与现代科学相融合奠定基础,对针刺疗法的临床应用和实验研究也具有一定的指导意义

The study focus on the neural electrical signals induced by acupuncture, analyze the cooperative action of the acupoints' structure and the associated chemical mediators during acupuncture, clarify the roadmap of the activation and transmission mechanism of acupuncture information through animal experimental design,signal analysis and mathematical models. First, test the peripheral neural electrical signals and the chemical mediators' (adenosine and histamine) concentration induced by acupuncture after acupoint pretreatment of collagenase, mast cell stabilizer and nerve blocker respectively, clarify the activation of acupoints structure participating in acupuncture (manual acupuncture and electro-acupuncture) and the source of associated chemical mediators in acupoints; Second, test the peripheral neural electrical signals induced by the injection of adenosine and histamine into acupoints respectively, compare them with those induced by acupuncture, and clarify these chemical mediators' regulatory function in acupoints; Third, test the neural electrical signals of spinal cord dorsal root induced by acupuncture and analyze the transmission of acupuncture information; Finally, establish mathematical models describing the acupuncture modulation effect and quantitatively analyze the response in acupoints to acupuncture. These work contribute to reveal the activation and transmission mechanism of neural signals induced by acupuncture from systems biology perspective, lay the foundation for the integration of acupuncture theory and modern science and further guide the clinical treatment and experimental research of acupuncture.
