脑梗死发生后脑侧支循环建立包括早期的血管舒张与随后的血管新生。一方面研究发现脑组织的RAS系统在脑血流的调节中具有双重效应,RAS除了促进血管收缩的经典ACE→AngII轴之外,还存在与其拮抗的ACE2→Ang 1-7轴;另一方面研究发现VEGF和Dll4/Notch信号传导途径在血管新生中扮演着重要的角色,本研究以MCAO大鼠为研究对象,以脑梗死后血管舒缩和血管新生为主线,从RAS系统的ACE- AngⅡ/ACE2- Ang 1-7轴和VEGF-Dll4/Notch通路的角度,应用激光多普勒、步态分析、激光共聚焦显微镜检测、实时荧光定量PCR、免疫组化、Western-blot酶联免疫等先进的技术和方法,动态直观观测大鼠脑梗死后血流改变、脑血管新生、神经功能缺损和上述通路关键指标的相关变化规律和针刺干预效应,探讨针刺人中穴对脑梗死后侧支循环的干预机制。
