作为一种重要的物质基本属性,磁性在药物研究领域已体现出巨大的应用潜力,而在天然药物化学方向亟待新思路进行开发。本课题以磁性和天然活性成分的结合为切入点,基于前期所建立的磁性衍生化方法,选择异喹啉类代表性生物碱单体作为模式分子,首先针对其磁性衍生物与分离密切相关的主要性质进行重点研究,并通过和原型分子以及彼此之间的详细对比掌握核心的结构-性质关系;进而在此基础上考察不同状态和分离模式下目标磁性生物碱单体及总碱部位受外磁场这一特殊环境影响所体现出的响应行为和分离趋势,探索有效的磁分离途径及其中关键因素的影响规律和内在机制,并同常规技术手段进行比较、验证;最后根据全部研究结果凝练具有系统性和实用性的基本理论及指导原则。本研究旨在促进天然药物化学和多个学科的结合,为建立成熟完善的磁分离方法完成必要的前期实验探索,通过丰富和补充既有研究手段为目前天然药物研究注入新的动力与活力。
英文摘要
As one of important basic nature of substance, magnetism has shown great application potential in pharmaceutical research. However, it urgently needs to be applied in the field of natural medicinal chemistry and new thinking is being expected. In view of this status, the study is based on the characters of common natural products and selects representative isoquinoline alkaloids as template molecules according to earlier research, which will be derivatized through simple procedure to be magnetic. The main properties and behaviors closely related with separation will be investigated in detail firstly. On the basis of the comprehensive comparison between magnetic alkaloids and contrast between them and prototype alkaloids, it will be explored and mastered about the key relationship between their structures and properties. According to the basic rules and fundamental data obtained from above studies, their magnetism will be applied in potential extraction and separation process, and magnetic responsiveness and separation trend of target alkaloids together with their bioactive fraction will be explored in various statuses and conditions. Then important influencing factors and mechanism in efficient separation mode are planned to be investigated through validation and comparison with conventional techniques. Finally, the related nuclear theories and guidance principles will be summarized from all the results and findings for the foundation of optimal, efficient and mature methodology in future. The study is aimed to propel the efficient combination of natural medicinal chemistry and related subjects, and provide necessary elementary exploration for the development of mature and perfect magnetic separation method, which is also hoped to inject new vitality into the present study of natural medicines as helpful supplement of current approaches.
