经临床观察、药效物质基础、体外骨重建培养体系的研究证实:土家族药-刺老苞根皮有效成分群不仅具有降糖、调脂、减轻胰岛素抵抗作用,而且可增强VEGF、TGF-β、FGF、PDGF、BMPs、ATF4及受体等相关细胞因子的分泌与表达,诱发骨周边血管生成,加快胶原合成与钙盐沉积、提高骨生物力学性能,促进骨形成,加快骨重建进程。基于上述工作基础,本课题旨在利用mirco-CT手段结合组织病理学、计算数学方法观测骨组织微结构,结合前期工作(青年基金项目、面上基金项目)较全面评价刺老苞根皮有效成分群对糖尿病骨重建的修复疗效。同时尝试性从IR-IRS-PI3K与Wnt/β-catenin信号耦联网络节点深入研究刺老苞根皮有效成分群对高糖状态下骨重建过程骨基质形成、间充质细胞分化诱导(骨诱导)、成骨与破骨细胞的趋化、增殖、分化的分子生物学机制。

The studies of clinical observation, osteoblast-osteoclast cell system in vitro and animal models in vivo confirmed that the active components of Aralia echinocaulis Hand. Mazz can not only lower blood sugar, regulae blood lipid and reduce insulin resistance, but also enhance secretion and expression of some correlated cytokines like VEGF, TGF-beta,FGF,PDGF,BMPs and ATF4, induce vascularization in callus, speed up collagen synthesis and calcareous deposit, improve the quality of callus biome, chanics and bone formation, which promote bone healing. Based on above-mentioned studies, in this project, we will observe 3D callus, microarchitecture by micro-CT, tissue pathology technique and computational mathematics methods, combined with the preliminary work (the project of National Natural Foundation) to comprehensive evaluate the repair efficacy of active components of Aralia echinocaulis Hand. Mazz on T2DM-OP (Type-2 Diabetes mellitus Osteoporosis). Meanwhile, we will study that the molecular biology mechanism of active components of Aralia echinocaulis Hand. Mazz in process of T2DM-OP through the key signaling molecules between IR-IRS-PI3K and Wnt/beta-catenin coupled network, and clarify the molecular biology mechanism of bone matrix formation, differentiation of mesenchymal cells (bone induction), chemotaxis, proliferation and differentiation of osteoblast and osteoclast during T2DM-OP healing process.
