成年卵巢由上千个不同发育阶段的卵泡组成,是正常机体极少数可发生周期性血管新生和重构的器官。卵泡发育及排卵是女性妊娠的基础,其过程和机制可能与卵巢局部的血管新生密切相关。本项目首次通过全组织透明染色技术,将完整卵巢透明化处理但保留组织内部精细的结构和生物信息分子,运用免疫组织化学染色技术对同一组织进行多轮不同抗原的特异性染色,通过激光共聚焦显微镜获取图像数据、计算机软件三维成像分析,直观描绘出(1)卵巢血管的三维空间分布和时空变化规律;阐明卵巢血管新生对卵泡发育和排卵的影响。(2)卵巢血管新生在PCOS卵泡发育和排卵障碍中的可能机制。(3)针刺通过对卵巢血管的调节对卵泡发育和排卵的影响。 这对卵泡发育的基础机制研究;离体或在体促进卵泡发育的临床实践;对大龄女性、PCOS、卵巢早衰及肿瘤放化疗术后有生育要求的女性的残存卵泡激活、发育和针刺治疗排卵障碍型不孕症等提供研究思路和科学依据。

Unique for the ovary, hormonally regulated folliculogenesis, follicle rupture, luteal formation/regression and associated vasculature changes lead to tissue remodeling during each reproductive cycle. Follicular development and ovulation is the basis of female pregnancy. The process and mechanism of follicular development and ovulation is highly related with local vascular and ovarian periodic reconstruction. In the present study, for the first time, we turn the whole tissue transparent (Clarity), but retain internal fine structure and molecular biological information. Then immune-staining of multi-round specific markers will be used to show 3D imagings obtained by laser scanning confocal microscope. Using the Clarity approach and marker immunostaining, we identified individual follicles and local vasculature network in intact ovaries: (1) to understand vividly the internal vascular network 3D distribution and spatiotemporal principles, blood vessels trends, estrous cycle dynamic, the relationship of angiogenesis and follicular development will be presented; (2) to study the abnormal vascular function in polycystic ovary syndrome (PCOS) – one typical disorder of follicle development and ovulation; (3) illuminate the effects of acupuncture on follicle development and ovulation by regulating angiogenesis of local ovarian vasculature. This study helps to understand the essential roles of vasculature in organizing follicles; promote in vitro and in vivo activation of follicular development; provide the fertility possibility for middle-age women, women with PCOS, premature ovarian failure (POF) and tumor radiotherapy and chemotherapy; develop scientific research and clinical practice of acupuncture in the treatment of anovulatory infertility.
