姜为最常用的中药和调味料之一。长期临床实践和现代研究证实,姜具有明显的防治脂糖代谢紊乱的作用,与其调理脾胃以防痰湿、调控营养物质的转输紧密相关,但其中的分子事件尚不清楚。脂肪酸转运酶FAT/CD36介导对脂肪酸的转运,与脂糖代谢密切相关;其在骨骼肌细胞的亚细胞分布决定着脂肪酸摄取、储存和利用平衡。我们前期研究表明,姜提取物及其主要代表活性成分[6]-姜辣素在改善脂糖代谢紊乱中,影响了骨骼肌细胞FAT/CD36的亚细胞分布。基于此,本课题紧紧围绕“FAT/CD36“这一调控“重要营养分子---脂肪酸”转输的关键因子,以骨骼肌“FAT/CD36的蛋白质翻译后修饰---亚细胞定位---脂肪酸摄取、储存和利用平衡”这一连锁反应链条为主线,揭秘姜改善脂糖代谢紊乱的潜在机制,为姜“调理脾胃”,防治代谢性疾病提供现代分子生物学依据,也为中医脾胃实质及其它调理脾胃方药防治痰湿证的研究提供科学依据和新思路。

英文摘要

Ginger is one of the most commonly used spices and medicinal plants around the world. Ginger is attributed to spleen and stomach channels, belongs to a spleen-stomach-regulating herb, and is widely used for the prevention and/or treatment of Phlegm-dampness syndrome in TCM. It has been well demonstrated that ginger improves numerous metabolic disorders. However, the underlying molecular mechanisms are still unclear. Fatty acid translocase (FAT/CD36) is closely associated with lipid and glucose metabolism. Fatty acid uptake, store and oxidation are highly regulated by FAT/CD36 at several subcellular sites in skeletal muscle fibers. Our research team has found that ginger and its prominent active component [6]-gingerol improve fructose overconsumption-induced fatty liver, hypertriglyceridemia and insulin resistance via subcellular redistribution of FAT/CD36 in skeletal muscle fibers. Based on this important finding, the proposed project intends to investigate how ginger and [6]-gingerol regulate FAT/CD36 protein posttranslation modification----subcellular location----fatty acid uptake, store and oxidation. The findings of this project will not only supply with molecular basis for ginger to improve metabolic disorders, but also provide new insight into the elucidation of the spleen and stomach-regulating nature in TCM. Thus, this project has novelty and significance.
