免疫治疗是胃癌综合治疗模式的重要组成部分,提高胃癌疫苗的疗效是胃癌临床的迫切需求。本项目在申请人前期工作发现“红细胞膜纳米粒子-水凝胶复合药物投递系统”具有靶向性、缓释性、持续性优势的基础上,建立以肿瘤细胞膜为载体,靶向DC的纳米化全细胞疫苗,联合局部清除肿瘤相关巨噬细胞的白蛋白-聚乙二醇载药可注射水凝胶的投递系统,研究其增强肿瘤疫苗的治疗效果及作用机制。着重研究该体系在提高抗原递呈效率,调节肿瘤免疫抑制微环境,促进免疫效应细胞应答等方面的优势,进一步深入挖掘胃癌疫苗的临床应用潜力,为胃癌的治疗提供新型、高效的治疗手段,为新型药物投递系统的开发和临床转化提供科学依据。

Immunotherapy is an important part of comprehensive treatment of gastric cancer. It is urgently needed to improve the efficacy of gastric cancer vaccine in clinical. In the previous work, we have investigated the targeting, controlled-release and sustainability of erythrosome-hydrogel complex drug delivery system. This project aims to fabricate the delivery system composed of DC-targeting nano-whole-cell vaccine carried by tumor cell membrane and tumor-associated macrophages eliminating BSA-PEG injectable hydrogel, and the treatment efficacy as well as mechanism of tumor vaccine will be evaluated. The project focuses on the advantage of this system in antigen presentation, tumor immune suppressive environment regulation and immune cell responses promotion. We hope that this program will explore the potential of gastric cancer vaccine in clinical application, provide not only new treatment of gastric cancer, but also the scientific evidence to develop novel drug delivery system and transit them to clinical application.
