民猪是我国唯一适应了东北高寒气候的中国地方猪种,是国家级保护猪种。随着季节的变化,着生和脱落绒毛是其抗寒性的主要表现之一。目前尚未见到有关猪绒毛生长规律及分子机制方面系统研究的报道。本研究以具有绒毛的民猪(二民猪、荷包猪)为实验材料,通过绒毛生长期、退行期、休止期三个时期的观察、检测,分析民猪绒毛季节性变化规律;利用高通量测序技术,在转录组水平、启动子甲基化情况及相关转录因子表达水平等方面鉴定绒毛发育和季节性变化的候选基因,采用生物信息学的方法对测序结果进行分析,对获得的差异基因进行通路富集,以确定民猪绒毛季节性生长的分子调控通路和在绒毛发育过程中具有重要作用的候选基因及作用机制。阐明民猪绒毛季节性变化的分子机制,为揭示民猪抗寒特性的分子机理提供基础,为解决人类毛发再生难题提供有效参考。

Pig is the only Chinese local pig adapted to the northeast cold climate of China. With the change of the seasons, the birth and the shedding of the hair is one of the main manifestations of the cold resistance. At present, there has not been a systematic study on the growth pattern and molecular mechanism of the growth of the pig. In this study, with the villi of Min pig (Er Min pig, Hebao pigs) as the experimental material, through the hair growth period, catagen and telogen three periods of observation, detection, analysis of minpig villi seasonal changes; the use of high-throughput sequencing technology at the transcriptional level, promoter methylation and the related transcription factor expression level and identification of hair growth and seasonal variation of the candidate gene, using the methods of bioinformatics analysis of gene sequencing results were obtained, the difference of the pathway enrichment, to determine the seasonal growth of minpig villi molecular regulatory pathway and play an important role in the development process of candidate genes in the villi and mechanism of action. To elucidate the molecular mechanism of seasonal variation in the seasonal variation of the Min pig"s hair, to provide the basis for revealing the molecular mechanism of cold resistance characteristics of the Min pig and to provide an effective reference for solving the problem of human hair regeneration.
