青礞石对癫痫疾病临床疗效确切，其化学成分为主要为金属元素且主要以离子形式存在。癫痫的发病机制与细胞内外离子浓度、离子通道、金属蛋白/酶的活性密切相关。鉴于前期研究，根据青礞石化学组成特点，基于癫痫的发病机制，提出其作用机制可能是通过金属离子与体内相关金属蛋白/酶活性中心结合，而干预调控细胞内外金属离子的浓度和输运，通过影响细胞通路或离子通道中的一组或几组金属蛋白/酶而减缓神经细胞去极化反应及爆发性放电发挥作用。其进入体内各种金属离子的相对含量、价态、形态及与金属蛋白/酶的结合状态为其发挥效应的关键因素。本课题利用金属组学相关理论和方法，整合多学科交叉研究思路，研究其在动物体内金属元素的分布、相对浓度、价态等，鉴定组织中差异金属蛋白，明确差异金属蛋白的结构、与金属离子的结合位点及其功能和生物效应，并与药效指标相关联，揭示其对癫痫作用的效应本质。本课题为加速矿物类中药的基础研究进程提供了思路。

Chloriti Lapis(CL) used in Chinese medicine in treatmeng of epilepsy is effective which chemical composition are inorganic elements, and mainly exists in the ion form. The pathogenesis of epilepsy are closely related ion concentrations inside and outside cells, ion channel and metalloproteinase activity. On the basis of preliminary studies and according to the chemical composition characteristics of CL and the pathogenesis of epilepsy and phleg, We proposed that the CL mechanism in treatment of epilepsy may be through a combination of metal ions in vivo and metalprotein/metalenzyme activity center and intervention regulation of intracellular and extracellular metal ion concentration and transport, by influence cellular pathways and ion channel of a group or several groups of metalprotein/metalenzyme and slow down the neural cell polarization reaction and explosive discharge to play a role. CL inside the body of various ions relative content, valence, and protein-bound state is the key aspect of its display effect. This subjects using metallomics theories and methods, integration of multidisciplinary research ideas, to study CL in animals of metal elements in the distribution and relative concentration, valence,and so on. Identification of biological tissue in different metal protein, clear different metal protein structure, and the metal ion combined with site, function and biological effect, and associated with pharmacodynamic index, to reveal the nature of the effect of CL in treatment of epilepsy disease in clilic.
