研究认为阿尔茨海默病(AD)与脑组织线粒体能量代谢异常密切相关。针刺能影响线粒体能量代谢相关功能,但分子机制不清。定位于线粒体膜上的跨膜转位因子复合体(Translocases)是线粒体前体蛋白进入线粒体内的通道和关键因素,对于维持线粒体能量代谢功能起重要作用。基于前期研究基础,我们认为针刺治疗AD与线粒体能量代谢相关,可能是通过激活跨膜转位因子复合体来实现的。本课题以SAMP8小鼠为研究对象,采用透射电镜、实时荧光定量RT-PCR、Western blot等方法,观察针刺对神经元线粒体跨膜转位因子复合体的活化及Tom40、Tim20和Tim23蛋白基因表达的影响,通过深入研究针刺对线粒体跨膜转位因子复合体的作用,探讨针刺调节神经元线粒体能量代谢的作用机理。本研究对阐明针刺改善线粒体功能的分子机制,阐述针刺改善认知功能的机理有重要意义,也能为针刺治疗AD提供坚实的理论依据和实验依据。

Research suggest that Alzheimer's disease (AD) is closely related with energy metabolic disorder of the mitochondria in the brain. Acupuncture can affect the function of the mitochondrial energy metabolism, but the molecular mechanism is not clear. The translocases localized to the mitochondrial membrane, is the key channel of the mitochondrial precursor protein into mitochondria, and plays a vital role in maintaining mitochondrial energy metabolism. Based on the previous research, we believe that the effects of acupuncture on AD are associated with the mitochondrial energy metabolism, which is may be implied through the activation of translocases. We will select the Senescence Accelerated Mouse P8 (SAMP8) as model to observe the effects of acupuncture on the activation of the membrane translocation factor complex, and the Tom40, Tom20 and Tim23 protein and the mRNA expression levels of neuron mitochondria, using transmission electron microscope, real-time RT-PCR technique and Western blot methods. We will further study the effects of acupuncture on the translocases to explore the mechanism of acupuncture in regulating the neuron mitochondrial energy metabolism. This study is of great significance to clarify the molecular mechanism of acupuncture in improving mitochondrial function, and to expound the mechanism of acupuncture in improving the cognitive function. It is can also provide solid theoretical and experimental evidence for acupuncture in treating AD.
