放置宫内节育器(IUD)是国际普遍使用女性避孕法,我国是IUD使用最多国家,但IUD出血副反应是计生领域研究难点。课题组前期证实IUD避孕大鼠子宫内膜血管平滑肌(VSMC)存在表型异常,宫宁颗粒可使内膜VSMC合成型转化为收缩型;目前公认Rho/ROCK通路是VSMC表型调控重要环节。基于此,课题提出:子宫内膜VSMC表型异常是发病关键环节,宫宁颗粒(三七茜草)有效组分可能通过Rho/ROCK通路逆转血管病理性重塑而发挥止血。本研究将建立无创IUD避孕大鼠和体外人子宫内膜VSMC模型,采用透视电镜、Western Blot、qPCR和Rho/ROCK阻断剂等,观察三七茜草组分干预Rho/ROCK通路炎症因子、表型转化指标(a-sMA、sM22a、G-actin)、MLCP、RhoA/ROCK I及底物MBSThr-697磷酸化和增殖凋亡表达,从而明确治疗机理和丰富计生领域的中医药理论内涵。

Intrauterine device (IUD) is the international commonly used female contraceptive methods, the use of the IUD most countries, but the side effects are bleeding IUD birth control research in the field difficult. Our previous confirmed IUD contraceptive endometrial vascular smooth muscle (VSMC) presence of phenotypic abnormalities, endometrial Gongning particles can be converted to synthetic VSMC contractile phenotype; now recognized as Rho / ROCK pathway is an important part of the regulation of VSMC phenotype.
