PM2.5 急慢性暴露已经成为加重我国缺血性心脏病死亡率不断增加的潜在危险因素之一,目前缺乏有效的预防控制方法。我们的前期研究发现,活血解毒法、益气养阴法方药对PM2.5促发大鼠心肌I/R损伤有明确的保护作用。本课题在前期研究基础上,从损伤保护、功能改善两个方面构建防治PM2.5促发心肌缺血性损伤药效评价方法体系,通过小动物动态可视化技术、活细胞工作站、细胞内颗粒定位及染色技术、芯片技术、RNA干扰技术等系列方法,在整体、离体器官、细胞3个水平进一步确证、比较两种治则方药干预PM2.5促发心肌I/R损伤的药效作用,发现其作用的异同、特点、优势环节,并循Keap1-Nrf2-ARE信号通路对线粒体抗氧化功能的调控、细胞内吞颗粒启动NLRP3炎症体路径深入探讨两种治则方药作用机制,为相关治则在中医临床PM2.5促发心肌缺血性疾病防控中的应用以及新药研发提供数据支持。

Long- and short-term exposure to PM2.5, has become one of potential risk factors for the mortality of ischemic heart disease increasing in China, and there is a lack of effective prevention and therapeutic measures at present. Our previous studies shows that activating blood circulation and removing toxics prescription, supplementing Qi and nourishing Yin prescription are effective treatments for myocardial ischemia reperfusion injury triggered by PM2.5. Based on the previous research, this study from two aspects of injury protection and function improve-ment, intend to the construction of methods and technology system for the prevention and treatment of myocardial ischemic injurycontributed to PM2.5. Using series of methods, such as inverted microscope and a high-speed video camera system, Live Cell Imaging System, localization technology of intake particle, dyeing technology, chip technology and RNA interference technology, to confirm the therapeutic effects of two kinds of therapeutic formula? for myocardial I/R injury triggered by PM2.5 at all of the animal, organ and cell detective levels. and comparing the similarities and differences , characteristics and advantages between two kinds of therapy. The investigation on the mecha-nism focous on Keap1-Nrf2-ARE on the mitochondrial antioxidant function and Endocytosis-NLRP3 inflammation. We will provide evidences for clinical and R&D study of Traditional Chinese Medicine.
