本项目拟采用RT-PCR、Western-blot等分子生物学手段,针对前期研究中发现的三倍体雌性虹鳟性腺发育过程中卵母细胞败育并呈类雄性化的现象,研究其性腺发育的基因调控机制,特别是Cyp19a、Dmrt1、Foxl2、Sox9等性控基因的表达,并应用外源激素对三倍体雌性虹鳟诱导和回救,佐证Cyp19a信号通路的作用;通过泛素连接酶3底物受体基因DDB1的克隆与表达以及自噬标记蛋白LC3的表达,研究三倍体雌性虹鳟卵泡败育后的自噬特点,解析三倍体虹鳟卵巢发育中经历的分化-去分化-再分化(类雄性化)的特殊现象。研究结果能够丰富多倍体鱼类分子遗传学理论,为鱼类性别控制和育性控制育种提供科学的理论依据和安全有效的方法,以提高虹鳟群体产量、产品质量和经济效益。三倍体雌性虹鳟性腺分化与发育分子调控机制的研究国内外尚未见报道。

In this project, aiming at the discovery in previous study that oocyte of female rainbow trout of the triploid aborted and presented class male biological during gonad dysplasia, we study its gonad dysplasia' s gene regulation mechanism, especially Cyp 19a, Dmrt1, Fox12, Sox9 and other sex control genes' expression, using RT- PCR, Western- blot and other molecular biology methods, as well as induce and retrieval female rainbow trout of the triploid applying exogenous hormone to corroborating the effect of Cyp 19a signaling. In the research of clone and expression of ubiquitin ligases 3 substrate receptor gene DDB1 and the expression of autophagy- mark proteins LC3, we study the traits of follicle aborting during autophagy of female rainbow trout of the triploid, to parse the special phenomena that female rainbow trout of the triploid experienced differentiation-- dedifferentiation-- re differentiation( class male biology ) during ovarian development. The research results enriches molecular genetic knowledge of polyploid fish, provides the theory basis and effective and secure method for gender- controlling of fish and fertility controlling breeding, to increase population yield, products' quality and economic benefit of female rainbow trout. The research of gonadal differentiation and developmental molecular regulatory mechanism of female rainbow trout of the triploid has not been studied and set up at home and abroad.
