榕树和传粉榕小蜂间形成了一种十分独特且专一性很强的共生关系,也构成了一种典型的协同进化和物种共形成模式系统。研究发现线虫是该共生体系中的新成员。但还不知道线虫与传粉榕小蜂或榕树之间的相互作用关系特别是进化关系。应用分子系统发育分析方法研究线虫与昆虫之间的进化关系才刚刚开始,但目前尚未涉及线虫和榕小蜂间的分子进化关系方面的研究。本项目结合形态学和分子生物学特征鉴定榕树与榕小蜂共生体系中线虫和榕小蜂种类, 在此基础上试图通过分子系统发育分析途径,基于线虫核糖体和线粒体DNA序列,采用ClustalW、ModelTest, MrBayes、PAUP、TreeMap和 Treefitter软件,构建线虫和榕小蜂的分子系统发育树,进行拓朴学分析比较,探索线虫和传粉榕小蜂物种共形成问题,揭示两者间的分子进化关系,为深入研究相关线虫的多样性,特别是线虫和榕小蜂之间的相互作用及进化的分子机制提供理论基础。
英文摘要
The relationship between figs and pollinating fig wasps is not only a very unique and highly specific symbiotic, but also a typical co-evolution and co-speciation model system. The studies have found that nematodes have become a new component of the symbiotic system. However, there is no idea about the interaction especially evolutionary relationships between nematodes and pollinating fig wasps or figs. Studying on evolutionary relationships between nematodes and insects using way of molecular phylogenetic analysis has only just begun, but it is not yet going through into the molecular evolutionary relationships between nematodes and pollinating fig wasps or figs. In this project combined morphological and molecular characteristics of the nematodes and pollinating fig wasps will be employed to identify species of nematodes and wasps in the mutualism. On the basis of species identification, phylogenetic trees inferred from ribosomal and mitochondrial DNA sequences of nematode and wasp will be constructed by using ClustalW, ModelTest, MrBayes, PAUP, TreeMap and Treefitter softwares, to explore co-speciation of nematodes and wasps, and further to reveal molecular evolutionary relationships between them based topology analysis. This should be helpful to further study on nematode diversity, and particularly to provide the theoretical foundation for study on the mechanisms of interaction of nematodes and wasps, and molecular evolution.
