对鹅掌楸保存种质资源以及鹅掌楸变异类型"北抗"鹅掌楸等进行采样,同步进行抗寒生理生化指标测定和多个表型性状调查测定,提取抗寒变异类型与极易受寒个体的RNA,进行转录组测序,进行比较和寻找抗寒通路,发掘相关的抗寒基因,进一步筛选SNP位点,在提取更多样品进行DNA测序基础上,寻找到足够区分鹅掌楸种质样品的有效SNP位点,建立鹅掌楸种内群体及变异类型的SNP指纹图谱库。将鹅掌楸种质资源关键表型性状、生理生化指标和SNP标记的基因频率建立关联分析,对人工保存种质资源进行综合评价,并研制鹅掌楸种质资源快速鉴定技术体系。在此基础上,揭示鹅掌楸种质资源的遗传多样性、遗传关系、评价鹅掌楸种质资源遗传保存效果和可靠性,提出鹅掌楸种质资源的安全保存与合理利用策略。
英文摘要
Genotypical identification of important characteristics of forest genetic resources is of significant importance to explore potential uses of genetic resources. Liriodendron chinense is an important native plant in southern China with fiber and ornamental values. Many people hope to introduce it to northern China. But currently the genetic background of its cold resistance and other characteristics is not clear. And genetic relationships among populations and individuals are not studied. This study intends to find relative genes of cold resistance and employ SNP markers to survey the genetic diversity and patterns of variation of Liriodendron chinense and its relationships among populations and individuals within species. DNA will be extracted from samples taken from the natural range and the planted trees for SNP analysis. Based on the SNP markers analyses, a fingerprint library of the molecular markers will be constructed. The information generated from the genetic survey of SNP markers, will be used to understand the genetic diversity, genetic structure of planted populations, and attempts will be made to identify unique genes or frequencies of populations. Moreover, the SNP markers will be further used for genotypic identification of key phenotypes. There will be a great potential to apply the expected outputs from the project in genetypical identification of key phenotypes of the genetic resources.
