本研究根据贾第虫这一双核原生动物在体外可反复传代培养呈现“永生化”现象以及端粒酶与细胞永生化密切相关的理论,从贾第虫端粒酶GdTERT相关蛋白为切入点,利用酵母双杂交技术筛选GdTERT相关基因蛋白,通过双分子荧光互补技术确定其在贾第虫体内互作及在双核中的定位,采用基因转染和敲除技术确定GdTERT相关基因蛋白在端粒酶活性调节中的作用,从而为揭示贾第虫端粒酶活性调节机制、双核功能、“永生化”现象或生存策略提供试验数据。同时也为贾第虫病防治提供新的药物和疫苗作用新靶位, 因而具有重要的理论意义和应用前景。
英文摘要
According to immortalized phenomenon of protozoan Giardia contained two nuclei which can be passaged repeatedly in vitro and theory of close relation between telomerase and cell immortalization, GdTERT associated protein will be studied.The GdTERT associated proteins will be screened using yeast two hybrid. By bimolecular fluorescence complementation technology, the interaction of among proteins and localization in two nucleuses of Giardia will be determined. The regulated telomerase activity of proteins will be identificated using gene transfection and knockout technique. It will provide experimental data to clarify the regulation mechanism of telomerase activity and reveal the function of two nucleuses,immortalized phenomenon or survival strategy of Giardia. Also, it will provided new drug and vaccine targets against Giardia. So it has important theoretical significance and application prospect.
