大气CO2浓度升高越来越被人类关注。本项目拟采用CO2人工气候箱设置370 ppm(低)和750 ppm(高)2个CO2浓度,探讨CO2浓度升高和Wolbachia感染互作对农田优势种蜘蛛草间钻头蛛的生殖、适合度、捕食功能反应、体内营养物质含量以及消化酶、保护酶和解毒酶活性等生理、生化特性的影响;利用高通量测序技术测定不同CO2浓度和Wolbachia感染与否的4个处理组合下草间钻头蛛的转录组,找到不同处理组合下差异表达的基因,克隆获得引起响应的候选基因,通过qRT-PCR和Western blot检测候选基因的表达和调控,揭示草间钻头蛛对CO2 浓度升高和Wolbachia感染响应的分子机制。本项目将为研究动物应对CO2浓度升高的响应机制开辟新的思路,丰富Wolbachia和CO2浓度升高在捕食性天敌领域的理论研究,为农田优势种蜘蛛的保护与利用供理论依据。

More and more people pay attention to the increase of atmospheric CO2 concentration. The effects of elevated CO2 concentration on the reproduction, fitness, predation, nutrient composition and enzymes(digestive enzymes, protective enzyme and detoxifying enzymes) activities of Hylyphantes graminicola will be investigated in climate chambers with two different concentrations of CO2 (low, 370 ppm and high, 750 ppm); The transcriptomes will be determined and the differentially expressed genes will be found from four treatment combinations of H. graminicola under two different CO2 concentrations and Wolbachia infection (un-infection) by using high-throughput sequencing. The candidate genes response for the interactions of elevated CO2 concentrations and Wolbachia infection will be cloned by RACE. The expression and regulation of candidate genes will be assessed by using qRT-PCR and Western blot to reveal the molecular mechanism of H. graminicola responding to the elevated CO2 concentration and Wolbachia infection..This project will open up new ideas for studying the mechanism of animals responding to elevated CO2 and will enrich the theoretical researches of how Wolbachia and elevated CO2 concentration impacting on the predatory natural enemies. It will also provide the theoretical basis for the protection and utilization of dominant species of spiders in the farmlands.
