多囊卵巢综合征(PCOS)是一种生殖及内分泌系统障碍并存的临床综合征,已成为临床上造成女性不孕症的主要原因之一。尽管有证据表明,小檗碱对于PCOS有良好的治疗效果,但其作用机理仍不清楚。本项目在前期小檗碱激活AMPK信号通路改善胰岛素抵抗研究的基础上,分别以丙酸睾酮诱导的PCOS大鼠和高糖高脂诱导的人卵巢颗粒细胞系为研究对象,应用免疫组化、Western blot、RT-PCR等技术,观察小檗碱对糖脂代谢、性激素水平、卵泡发育情况以及卵巢LKB1-AMPK-MAPK3/1信号通路相关分子表达的影响,验证小檗碱改善胰岛素抵抗、性激素分泌和促进卵泡发育的多重效应,为小檗碱治疗PCOS提供理论依据,也为中医药在生殖医学的推广及应用奠定基础。

Polycystic ovary syndrome (PCOS) is a reproductive-metabolic disorder which has become one of the main cause of female infertility. Although evidence suggests that the effect of Berberine(BBR) has a positive therapeutic effect on PCOS, its exact mechanism of action remains unclear. Basing on the previous study about BBR activates AMPK pathway to improve insulin resistance, this project will adopt Testosterone propionate-induced PCOS rats and high glucose and fat-induced KGN cells to observe the effect of BBR on the glucose, lipid, sex hormone levels in serum and the condition of follicular development,and to detect the expression of LKB1-AMPK-MAPK3/1 signaling pathways related molecule usingimmunohistochemistry, western blot, RT-PCR. The objective is to verify that BBR has multipath effects include improving the insulin resistance, sex hormones secretion and promoting the follicular development through influence the expression of LKB1-AMPK-MAPK3/1 signaling pathways. It can provide theory evidence for BBR in the treatment of PCOS, and establish the foundation of the application and extension of TCM in reproduction medicine.
