增液汤是临床中医治疗阴虚热盛型糖尿病(2型糖尿病早期)的常用方剂,由玄参、生地和麦冬三味常用滋阴药组成,其降糖疗效及可能机制也有较多文献报道。然而,由于中药方剂化学复杂性和生物多效性的特点,导致增液汤的物质基础和组方原理依然不明。网络分析技术在分析、整合和总结非线性复杂数据方面的优势,使其在中药物质基础研究中具有良好的前景。本研究拟以增液汤为研究对象,对其治疗糖尿病的物质基础进行研究: 通过引入“效应”这一要素,对现有的网络分析方法技术加以改进;采用多个学科的方法技术,结合增液汤及其组成药材的体内过程研究和效应研究,针对增液汤及其组成药材分别构建“成分-靶点-效应”复合网络体系;预测与评估增液汤及其组成药材的物质基础,作用靶点和相关效应,并结合实验结果对网络分析结果进行简化与验证,据此阐明增液汤组方原理的科学内涵,并建立一种基于网络分析的中药方剂物质基础和组方原理研究新模式。

Zeng-Ye-Tang is used to treat diabetes caused by extreme heat with yin asthenia (type 2 diabetes early) by traditional Chinese physician in clinic, which is composed of three traditional Chinese medicines (TCMs) for nourishing yin, Radix Scrophulariae, dried Radix Rehmanniae and Radix Ophiopogonis. Many pharmacological studies reported hypoglycemic effect and potential hypoglycemic mechanism of Zeng-Ye-Tang. However, due to the chemical composition complexity and biological pleiotropic characteristics of TCM prescription, the material basis and combination principles of Zeng-Ye-Tang is still unknown. Network analysis techniques have a good prospect in the material basis of TCM research because of its advantages of analyzing, integrating and summarizing nonlinear complex data areas. This study intends to study the material basis of Zeng-Ye-Tang for the treatment of diabetes: to improve the present network analysis techniques by the introduction of the element of "effect"; based on studies of effect and in vivo analysis for Zeng-Ye-Tang and its composition of medicinal herbs, and using approaches and technologies of multiple disciplinary, to construct "ingredient - target - effect" network of Zeng-Ye-Tang and its composition of medicinal herbs; to predict and to evaluate material basis, the role of targets and related effects of Zeng-Ye-Tang and its composition of medicinal herb according to network analysis results, which can be further simplified and verified based on the experimental results. These results can help clarify the scientific connotation contained in combination principles of Zeng-Ye-Tang, and this study will establish a new mode to be used in material basis and combination principles research of TCM prescription using network analysis.
