宫颈癌是最常见的妇科恶性肿瘤之一,难治性、复发性宫颈癌的早期诊断及治疗是宫颈癌诊疗的难点。近年来,小分子靶向多肽因其自身特点成为一种理想的肿瘤转移示踪及治疗的载体。随着分子超声造影技术不断成熟,靶向微气泡的应用成为对宫颈癌有效示踪及治疗的突破口。因此,本研究从前期研究所筛选的高转移肿瘤靶向多肽TMTP1入手,通过制备携TMTP1靶向多肽微气泡造影剂,采用分子超声成像技术对宫颈癌原发灶及转移灶进行活体示踪及成像;并通过超声波介导微泡造影剂破坏技术,将前期研究所获得、具有促进肿瘤细胞凋亡作用的TMTP1-TAT-NBD毒性多肽靶向转染入宫颈癌病灶,联合分析TMTP1-TAT-NBD毒性多肽对宫颈癌治疗作用价值。本研究拟为宫颈癌的诊断开辟新的思路及方式,为复发及难治性宫颈癌的靶向治疗提出新的有效途径。

Cervical cancer is one of the most common gynecological cancers. Early diagnosis and treatment of endogenous cervical cancer and refractory cervical cancer is a difficulty in the diagnosis and treatment of cervical cancer. In recent years, small molecules targeting peptide becomes an ideal tumor metastasis tracer and treatment carrier due to its characteristics. With the development of molecular ultrasound imaging technology continues to mature, application of targeted microbubbles becomes a breakthrough of effective tracer and therapy for cervical cancer. Therefore, this study starts with the high metastatic tumor target polypeptide TMTP1 selected from early research, through the preparation of microbubble contrast agent containing targeting peptide TMTP1, imaging and tracking the tumor and metastasis of cervical cancer by ultrasound molecular imaging techniques. Using ultrasound-mediated microbubble destruction(UMMD) technique, TMTP1-TAT-NBD toxic polypeptide compound,which is obtained in previous research and able to promote tumor cell's apoptosis,is transfected into cervical cancer and metastasis,to analysis the value of TMTP1-TAT-NBD toxic polypeptide compound 's therapeutic effect on cervical cancer .This study is supposed to discover new ideas and ways for diagnosis of cervical cancer, putting forward a novel and effective method for relapsed and refractory cervical cancer targeted therapy.
