大脑中枢系统疾病的药物治疗效果往往由于血脑屏障的存在而大打折扣,如何使用一种安全有效的载体,既不损害脑组织的正常机能,又能增加其在血脑屏障的通过率就显得尤为重要。我们采用具有良好生物相容性的新型DNA折纸纳米载体,以脑毛细血管内皮细胞上有较高表达的低密度脂蛋白受体为靶点,在不同形状DNA纳米载体上表征该受体的特异性配体ApoE。同时在该体系上耦连具有长波长发射光的上转化染料,制备成集靶向与成像为一体的多功能纳米载体。采用光学分子影像对不同形状DNA折纸载体的跨血脑屏障的效果进行在体、实时评估,从而筛选出最佳的纳米载体的形状,为后续中枢系统疾病的靶向递药打下坚实的基础。

Drug treatments of central nervous system diseases often compromised due to the presence of blood-brain barrier, and how to use a safe and effective carrier, neither damage the normal function of the brain, but also increase the permeability in the blood-brain barrier becomes particularly important. We use novel DNA origami biocompatible nanocarriers,which target the brain capillary endothelial cells high expression density lipoprotein receptor. The different shapes nanocarriers are modified with ApoE protein, meanwhile long emission wavelength dye is coupled on the surface. These multifunctional nanocarriers integrate targeting and imaging together. Optical molecular imaging is used to give different DNA origami shapes carriers acrossing the blood-brain barrier effeciency in vivo, real-time assessment. All abvoe lays a solid foundation for targeted follow-central nervous system diseases drug delivery.
