基于课题组前期系列研究发现:中医益气活血通络法防治脑缺血再灌注损伤(CIRI)疗效肯定,可通过调控AktmRNA/蛋白及其相关凋亡因子的表达,保护受损神经细胞、促进神经功能恢复。本研究进一步从PI3K-Akt-mTOR-自噬轴通路,探讨该治法拮抗CIRI的可能机制。复制MCAO/R大鼠模型,应用慢病毒追踪、荧光成像、电镜、Western blot和Real-time PCR等方法,重点研究益气活血通络中药对脑缺血再灌注后自噬体、自噬流、自噬相关分子以及通路关键因子PI3K-p-Akt-mTORCI-p-P70S6K表达的影响:从PI3K-Akt-mTOR-自噬轴的分子作用途径深入阐释益气活血通络中药拮抗脑缺血再灌注后神经损伤的有效作用靶点。为临床研制出疗效显著、具有自主知识产权的治疗缺血性脑血管病的中药复方提供全新的科学依据,进而丰富中医治则治法学内容。

Our previous studies found that : the method ,the TCM benefiting qi for activating blood circulation to free the collateral vessels theory treat cerebral ischemia and reperfusion injury (CIRI) ,is effective, and through regulating the expression of AktmRNA /protein and related apoptotic factors, this method protects the damaged nerve cells and promotes neurological recovery. This further study explores the PI3K-Akt-mTOR-autophagy axis pathway and tries to find the possible mechanisms of the antagonism of CIRI. The method copies the MCAO / R rats model, using lentivirus tracking, fluorescence imaging, electron microscopy ,Western blot and Real-time PCR methods, and focuses on how benefiting qi for activating blood circulation to free the collateral vessels theory of Chinese medicine influences the expression of autophagy, autophagy-related molecules and the key access factors PI3KI-p-Akt-mTORC1-p-P70S6K after cerebral ischemia and reperfusion. Further elaborate the potential valid targets of benefiting qi for activating blood circulation to free the collateral vessels theory medicine treats cerebral ischemia and reperfusion injury .This method provides new scientific evidence for clinic through developing a significant clinical traditional Chinese medicine with independent intellectual property rights which can effectively treat ischemic cerebrovascular disease, and then enriches the content of Chinese treatment rules.
