虽然生物大分子药物发展较快,但其递释系统的释药机理研究明显不足,而适时有效释放是生物大分子药物递释系统发挥高效作用的关键问题之一。本课题选择具有代表性的给药途径(局部注射给药、全身注射给药和粘膜给药)和代表性的生物大分子药物(蛋白多肽、抗体、siRNA、亚单位疫苗),深入系统地研究其递释系统的释药行为和释药过程等,揭示生物大分子药物高效递释系统的释药规律、影响因素、调控因素等,从而阐明其释药机制,通过对释药速度、释药部位、释药环境的整体调控,实现生物大分子药物高效递释系统的适时有效释放,为设计新一代生物大分子药物高效递释系统打下良好的基础,有效促进生物大分子药物高效递释系统的发展,具有重要的科学意义和现实意义。本课题是本项目的重要分支和组成部分,本课题的研究将为项目研究任务的完成做出直接和重要的贡献。本团队研究力量较强、前期工作基础较好、研究条件具备,完成研究任务的可行性较强。
英文摘要
Despite the fast development of biomacromolecular drugs, there are obviously in lack of studies on the release mechanism of their drug delivery systems. However, the timely effective release remains one of the key issues for the efficacy of biomacromolecular drug delivery systems (BDDS). We choose representative delivery systems such as local injectable, intravenous injectable and mucosa delivery systems, as well as representative biomacromolecules including protein and peptide, antibody, siRNA and vaccine, to elucidate their release features such as release rules, impact and modulating factors, and to interpret the release mechanisms. We aim to achieve the timely effective release of drug from BDDS via modulation of release rate, position and microenvironment, which may build the foundation for the design of new-generation BDDS and promote the development of BDDS. As an important part and branch of the project, this study will contribute significantly to the accomplishment of the project. It is very likely that we could achieve our goal since we have good research team, previous preparation and research facilities.
