精原干细胞(SSC)是肾中所藏先天之精的物质基础,近年发现,细胞周期蛋白D(cyclin D)在SSC增殖中发挥着重要作用,PI3K-Akt通路传递的信号上调cyclin D的表达,使细胞顺利通过G1期进入S期,产生增殖效应。前期研究表明,补肾填精中药—养精胶囊能够通过PI3K-Akt通路促进SSC增殖,另外,将携带有cyclin D干扰片段的腺病毒感染SSC发现增殖活性降低,所以课题组推测补肾填精中药可能是通过PI3K-Akt通路上调cyclin D的表达来调控SSC的增殖功能。本研究拟利用成年雄性小鼠、细胞实验及小干扰RNA技术,进一步研究补肾填精中药对cyclin D的表达、定位和上游启动子的影响,明确是否通过cyclin D促进SSC的增殖,进而阐明补肾填精中药调控SSC功能的分子机制。这对于揭示“肾藏精”的科学内涵,应用中医药治疗由少弱精症引起的男性不育具有重要的理论和现实意义。

Spermatogonia stem cell(SSC)is the material basis of kidney's congenital essence. It is recently found that cyclin D plays an important role in SSC’s proliferation. The expression of cyclin D is up-regulated through PI3K-Akt pathway. Cyclin D influences on G1 phase and impels the transition from G1 phase to S phase. Previously, we demonstrated that kidney reinforcing drug, Yangjing capsule, could stimulate SSC to proliferate through PI3K-Akt pathway. Furthermore, SSC infected by adenovirus carrying cyclin D small interference RNA shown low proliferating activity. Therefore, we hypothesized that kidney reinforcing medicine could stimulate proliferation of SSC through PI3K-Akt pathway by up-regulation of cyclin D. This study intends to use adult male mice, cell experiments, and small interfering RNA technology to further study the effect of kidney reinforcing drug on the expression , location and promotor of cyclin D. Then it can be identified that whether it is through the role of cyclin D to stimulate proliferation of SSC. The molecular mechanisms that kidney reinforcing medicine on SSC function would be clarified. In conclusion, the study would contribute to elaborate the scientific connotation of “kidney storing essence”, and use Chinese medicine to treat male infertility caused by oligoasthenotspermia.
