天然香豆素类化合物具有多种生理活性,在对螨类控制上效果明显,靶标独特,具有很好的开发研究价值。香豆素母环上的C-6和C-7位点结构及官能团变化与该类化合物的杀螨活性密切相关,是研究天然产物杀螨剂构效关系和筛选新的天然活性化合物的重要切入点。为揭示香豆素母环C-6和C-7位点对于提高杀螨活性的机制,准确快捷地筛选天然活性化合物,本研究以简单香豆素类化合物为材料,以Ca2+-ATPase靶标导向为目标,从生物测定、酶活评价和分子靶标三个层次来明确天然小分子香豆素类化合物杀螨活性的构效关系,比较及验证香豆素母环C-6和C-7部位不同小分子功能团的活性,在此基础上筛选靶标明确、活性突出、效果显著的植物性杀螨活性化合物,为进一步阐述香豆素类化合物生物活性多样性的机制,开发新型的天然产物杀螨剂提供依据。

With a variety of physiological activity, natural coumarin compounds have a good research and development value for its obvious effect and unique target on controlling mites. Closely related to the acaricidal activity of natural coumarin compounds, the structure and functional change of C-6 and C-7 loci on coumarin ring become an important point to study the Quantitative Structure - Activity Relationship (QSAR) of natural products acaricide and screen new natural active compounds. To reveal the mechanism of the function of C-6 and C-7 loci on improving acaricidal activity and accurately and efficiently screen natural active compounds, this study, took simple coumarin compounds as materials and Ca2+-ATPase target-oriented as goal, made clear the QSAR of natural small molecule coumarin compounds’ acaricidal activity from levels of biometrics, enzyme activity evaluation and molecular targets, and verified the activity of small molecule functional groups on C-6 and C-7 loci. Based on this, the botanical acaricidal active compounds, with clear target, prominent activity, distinguished effect, were screened. These researches provide a basis for further elaborating the mechanism of the diversity of biological activity of coumarin compounds and developing new natural products acaricides.
