糖脂代谢紊乱造成的肝肾损伤严重危害我国人民的健康,临床缺乏有效防治药物。本课题组前期研究发现:中药糖肾方能改善脂代谢紊乱及肝肾脂质沉积,保护肝肾功能,但作用机制有待阐明。我们假说:糖肾方可能通过纠正糖脂代谢通路(mTOR, NF-kB, TGF-β/Smad等)紊乱,发挥肝肾保护和治疗作用。本项目拟利用加速性高脂高胰岛素抵抗小鼠(CRP转基因db/db)模型结合临床和细胞研究,应用先进分子生物学技术和RNA-seq基因测序方法,研究糖肾方纠正糖脂代谢通路紊乱所致肝肾损伤机制,阐明糖肾方“肝肾同治”的物质和理论基础,丰富祖国医学“肝肾同源”的理论内涵。

Metabolic disorders of glucose and lipid can cause liver and kidney injuries, which are now the serious risk to the health of Chinese people. Moreover, there are lacks of successful medicines to prevent and treat metabolic disorders-induced injuries in diabetic patients. Investigation from our previous project reveals that Chinese herbal medicine – Tang Shen Formula (TSF) can regulate lipid metabolism, prevent lipid deposition from the liver and kidney, and improve function of the liver and kidney in diabetes. However the mechanism remains to be elucidated. We hypothesize that TSF can regulate signaling pathways involving in glucose and lipid metabolism such as mTOR, NF-kB and TGF-β/Smad etc to prevent and treat liver and kidney injuries in diabetes. In the current proposal, we plan to employ CRP-Tg db/db mice that display rapid development of hyperlipidemia and insulin resistance, as well as clinical trial and cell culture to investigate mechanisms how TSF regulate signaling pathways in glucose and lipid metabolism and prevent liver and kidney injuries in diabetes by applying advanced molecular technologies and RNA-seq method. The findings from the study will improve our understanding of how TSF can prevent and treat both liver and kidney injuries induced by glucose and lipid metabolic disorders. It can further reveal the real contents of some traditional Chinese medicine concept.
