乙型肝炎病毒(HBV)感染已经成为全球性危害的态势,抗乙肝病毒治疗亟待重视。苦参乌梅汤为我院长期临床治疗慢性乙型肝炎的经验方,前期研究工作发现苦参乌梅汤能够显著影响HBV转基因小鼠血清中HBsAg的表达。在HBV转基因小鼠体内具有抗HBV活性。目前对苦参乌梅汤抗HBV作用机制尚不清楚,因此我们提出假设:苦参乌梅汤是否通过干预宿主细胞代谢起到抗HBV作用。本项目以苦参乌梅汤作为实验药物,以HepG2.2.15细胞作为实验模型,拟建立和完善基于气相色谱-质谱联用技术(GC-MS)的细胞代谢组分析方法及具有统计意义的多变量代谢组学数据分析方法;采用代谢组学、蛋白质组学以及分子生物学相结合的方法,研究苦参乌梅汤干预影响宿主细胞代谢组的变化规律,发现其相关的代谢物以及所受影响的代谢途径;阐明苦参乌梅汤干预与宿主细胞代谢改变之间的关系;深入探讨苦参乌梅汤抗HBV作用机制。

Because hepatitis B virus (HBV) infection has become the trend of global hazard,Anti-HBV treatment needs urgent attention. Preliminary studies found that Kuhseng-Mume Decoction can significantly affect the expression of HBsAg in serum of HBV transgenic mice. But the anti-HBV effect mechanism of Kuhseng-Mume Decoction is unclear, which researches have rarely been reported. We will carry out the anti-HBV molecular mechanism studies in vitro with Kuhseng-Mume Decoctionas an experimental drug, and HepG2.2.15 cells as an experimental model. We assume that Kuhseng-Mume Decoction may play a role in anti HBV through the host cell metabolic intervention. We would intend to develop gas chromatography mass spectrometry(GC-MS) -based metabonomics method with multivariate data analysis for cultured cells, and then to investigate the metabolic responses of he host cell intervened with Kuhseng-Mume Decoction using metabonomics, proteomics and validate the results by a range of molecular biological assays. Therefore, this study will find the related metabolites and metabolic pathways, elucidating the relationship and further discussing anti HBV mechanism.
