卵泡是哺乳动物卵巢的组成及功能单元,决定了其中包含的雌性配子-卵母细胞的发生、生长及成熟,对于雌性生殖力的维持具有重要意义。卵泡发育及卵母细胞成熟一直是本领域的研究热点,但研究进展受限于体外研究手段。本项目中,申请者利用条件敲除小鼠模型,在卵母细胞中特异敲除CK2β基因,综合运用免疫荧光、免疫印迹、免疫组化、活细胞动态观察、底物筛选及补救实验等研究手段,研究CK2蛋白在小鼠卵泡发育和卵母细胞成熟过程中的功能。此项研究将有助于对哺乳动物卵巢中卵泡发育及卵母细胞成熟的深刻认识,为女性不孕症的治疗和辅助生殖技术的进步提供一定的理论支持。

Follicles are basic units of mammalian ovary and support enclosed female gametes-oocytes to survive, grow and mature, playing indispensable roles in female fertility. Folliculogenesis and oocyte maturation have been the research hotspots in reproductive biology, while the studies are limited by uneffective methods. In this study, we specifically delete the gene CK2β in mouse oocytes in vivo by conditional knockout technology, combining the technologies like immunofluorescence, immunoblotting, immunochemistry ,live oocyte imaging,substrate screening and rescue experiments to study the functions of CK2 in folliculogenesis and oocyte maturation. The study will help to deepen the views of.folliculogenesis and oocyte maturation in mammalian ovary, and will provide theoretical support for the treatment of female infertility and progress in assisted reproductive technology.
