前列腺素类化合物作为一种局部激素在哺乳动物的生殖过程及机体免疫调控过程中均发挥着重要作用,但其对子宫内膜上皮细胞Toll样受体(Toll-like receptor,TLR)家族蛋白介导的天然免疫调控机制尚不清楚。本课题主要目的是阐明PGE2和PGF2α对奶牛子宫内膜上皮细胞TLR2、TLR4及RP105参与的信号转导通路激活的调控机制,分析TLR信号转导通路的激活及RP105对奶牛子宫内膜上皮细胞分泌功能及PGE2和PGF2α表达的影响;研究PGE2和PGF2α对子宫内膜上皮细胞相关TLRs及RP105表达的影响;研究PGE2和PGF2α对奶牛子宫内膜上皮细胞TLRs介导的信号转导通路激活的影响及RP105在此过程中的作用。该课题的研究成果可为临床上合理应用前列腺素类药物防治奶牛子宫内膜炎性不孕症提供免疫学理论依据和实验基础,紧密结合内蒙古地区的生产实践,具有良好的临床应用前景。

Prostaglandins, a class of local hormones, have been shown to play important roles in female reproductive processes and in immunoregulation processes of mammalian species. But the mechanism of prostaglandins on regulation of innate immunity mediated by Toll-family proteins in endometrial epithelial cells has not been studied in detail. The aim of this study is to investigate the mechanism of prostaglandin E2 and F2α on regulation of TLR2, TLR4 and RP105 mediated signaling activation in bovine endometrial epithelial cells. Experiments were carried out to analyse the effect of TLR signaling activation and the role of RP105 on the secretion function and prostaglandin E2 and F2α expression in bovine endometrial epithelial cells. In addition, to investigate the effect of prostaglandin E2 and F2α on TLRs and RP105 expression, as well as TLR signaling activation and the fuction of RP105 in bovine endometrial epithelial cells. The results of this study will provide immunology theory and experimental fundamentals for clinical treatment of endometriun-caused infertility in dairy cows by using prostaglandin drugs, which suit the situation of Inner Mongolia and have capacious clinical application.
