本课题从可逆性调节血脑屏障(BBB)通透性、增强对脑胶质瘤细胞的主动靶向及抑制多药耐药三个角度出发,解决脑胶质瘤患者的治疗疗效差、生存期短、死亡率高等问题。项目首先利用具有“引经入脑”中药的有效部位促进机体BBB生理性可逆开放,提高脂质体在脑组织中分布机率;然后利用主动靶向抗耐药脂质体对脑胶质瘤细胞的特异性靶向结合,实现对肿瘤细胞的主动靶向;最后利用粉防己碱的抗多药耐药性质,提高脑胶质瘤细胞对化疗药物的敏感性,从而达到协同靶向的治疗目的。课题拟从“引经入脑”中药有效部位的可逆性调节BBB通透性及联合给药后脑胶质瘤细胞凋亡的Fas/FasL信号传导通路两方面探讨课题的相关作用机制。本课题以中医药理论为基础,结合现代主动靶向给药技术,可进一步完善中医药理论与现代制剂技术的有机结合,为脑部疾病提供新的治疗模式,为拓展中医药在临床抗肿瘤方面的应用提供新的理论依据。

The aim of this study is to resolve the problem of poor curative effect, short lifetime, and high mortality in the treatment of glioma from the promotion reversible opening of blood brain barrier (BBB), the active targeting of tumor cells and the inhibition of multidrug resistance (MDR). Firstly, the reversible opening of BBB would be regulated by the active fraction of “guiding medicine into brain” traditional Chinese medicine, and the distribution probability of drugs in brain tissue is increased accordingly; Secondly, The effect of active targeting to glioma cells will be achieved via the specific combination of the active targeting anti-multidrug resistance liposomes and the tuomor cells; Thirdly, the sensitivity of anti-tumor drugs is improved in virtue of the inhibiting MDR ability of tetrandrine. Finally, the synergic targeting effects are achieved by using the above methods. The study will discuss the related mechanisms from the reversible opening of BBB and the Fas/FasL apoptosis signaling pathways after combined administration. It is based on the theory of traditional Chinese medicine, and combines with modern technology of targeting drug delivery. It will further improve the incorporation between theory of traditional Chinese medicine and modern preparation technology, provide a new treatment model for glioma, and offer theoretical and experimental bases for the expending clinical application of traditional Chinese medicine.
