本课题拟基于中药化学、中药分析、药理学、代谢组学等多学科研究技术与手段,建立《中国药典》多基源中药钩藤化学物质基础与传统药效的关联性研究方法,为药典钩藤项下五个来源的植物用药科学合理性评价提供研究依据。研究主要包括四个方面:基于实验室自建“钩藤植物次生代谢产物全息数据库”与化学计量学分析相结合的化学物质基础分析;基于传统的功能主治,建立高血压和惊厥药理模型,进行五种植物的活性比较分析;基于代谢组学技术与药理活性分析,建立内源性物质作为药物药效辅助评价的方法;最后,通过灰色关联度分析法,建立化学成分群与活性之间的关系,阐明与传统药效直接相关的化学物质基础,及相关来源的钩藤。通过上述多基源中药钩藤与药效相关的质量评价研究,为“多基源入药是否合理的研究”提供研究思路与方法。
英文摘要
The project is planned to establish an approach to evaluate the correlation between the chemical basis and pharmacological activities of traditional Chinese medicine Uncariae Ramulus Cum Uncis (Gou-Teng) based on the multiple technologies, including natural products chemistry, analytical chemistry, pharmacology and metabonomics. The project will provide the basis for the scientific evaluation of the five available sources of Uncaria stem with hooks. The project is designed to investigate four aspects. Firstly, chemical basis analysis will be performed based on the holographic database of botanical secondary metabolites and chemometrics. Secondly, the pharmacological activity evaluation of the five herbal medicines will be performed based on the animal models according to the traditional functions. Thirdly, the associated evaluation method is developed based on the metabonomics and pharmacological activity evaluation. And, ultimately, the correlation analysis between the chemical basis and pharmacological activities is employed based on the GRAGM (gray relational analysis grade method), which elucidate the therapeutic basis and related herbal medicines. Based on the above researches of quality evaluation of multi-source TCMs, The project will provide a novel solution for the issue “multiple sources for TCM is reasonable or not” in a scientific way.
