证候科学内涵阐释对于具化中医个体诊疗与精准医学、促进中西医融合,极具价值。以冠心病痰证、瘀证等为代表的证候生物学基础研究在多个不同层次指标、研究技术等方面已取得显著进展,值得借鉴;然而对痰→瘀、瘀→痰等证-证间的交互特征、多个生物学基础指标间的相互作用关系整合不足,亟需反映互作关系、整体性的思路与方法。. 本研究立足证-证间内在关系和整体性特征,从系统科学角度提出“痰瘀-生物分子网络不同状态与功能表达”假设,拟开展冠心病证候横断面调查基础上的纵向研究,揭示痰→瘀、瘀→痰、痰瘀互结等交互演变规律,同步以基因与表观遗传组(mRNA)、蛋白组、代谢组等多重组学技术检测痰瘀相关的“脂质、血管、炎症介导”相互作用组学指标网络,应用高维矩阵、社会网络分析等建构痰瘀对应的生物分子网络及其功能表型,从关系、网络角度阐明证候现代科学内涵,同时探索建立一种体现相互作用、关系网络角度的证候基础研究方法。
英文摘要
Based on the intrinsic relationship and integral characteristics between syndromes, we propose the hypothesis that "Phlegm - blood stasis -biomolecular networks might have different statuses and functional expressions. The study is therefore initiated to carry out a longitudinal study on the coronary heart disease based on cross-sectional survey, in order to unravel the interactive evolution rule of syndromes of phlegm → blood stasis, blood stasis → phlegm, and intermin-gled phlegm and blood stasis. At the same time, multi-omics technologies combining genomics, epigenomics (mRNA), proteomics, metabolomics will be employed to measure the interactomics indicator network--phlegm and blood stasis-related "lipids, vascular, and inflammatory mediators". High-dimensional Matrix and Social Network Analysis will be used to construct the phlegm and blood stasis biomolecular networks and the functional phenotype, in order to illuminate the modern scientific connotation of TCM syndrome based on relationship and network. Meanwhile, our research might also contribute to explore a novel regime for the basic biological research of TCM syndrome characterized by inter-relationship and relational network.
