艾滋病感染后，患者CD4+T细胞功能逐渐受损，Thl/Th2失衡，常见腹泻等症状。腹泻与HIV激活人体天然免疫导致的微生物转位相关，可通过LPS/TLR4信号通路激活NF-κB导致多种炎症因子紊乱。益气健脾中药益艾康可稳定或减缓CD4+T细胞的降幅，抑制异常免疫激活并减轻腹泻症状，但机制不明。本课题在前期基础上提出“益气健脾法通过LPS/TLR4信号通路调控HIV/AIDS患者Th1/Th2平衡，从而调节患者免疫功能”的假说。本课题通过检测HIV/AIDS肺脾气虚证患者LPS/TLR4信号通路相关基因、蛋白及细胞因子及Th1/Th2，探讨该信号通路与免疫平衡的交互作用，构建HIV/AIDS“病-证-基因-信号通路”互作网络模型。通过体外实验探讨益气健脾中药通过LPS/TLR4信号通路对Th1/Th2免疫调控作用机制，并与临床试验对照印证，为揭示中药治疗艾滋病的可能机制和途径提供新的思路。

After HIV infection, CD4+T cell function of patient gradually damaged, Thl/Th2 imbalance,and the diarrhea is common symptoms.They are correlation between microbial transposition and diarrhea, and HIV activates the NF-κB lead to disorders of inflammatory factor through the LPS/TLR4 signaling pathways. Yi-ai-kang can stabilize and slow decline of CD4+T cells,and inhibit abnormal immune activation, and relieve diarrhea.But so far, the mechanism is not clear.This study posit that strengthening spleen and nourishing Qi method adjusts Th1/Th2 balance based on LPS/TLR4 signaling pathway with HIV/AIDS patients.In this study,we will detection genes, protein and cytokines related LPS/TLR4 signaling pathways and Th1/Th2 with HIV/AIDS strengthening spleen and nourishing Qi patients.To explore the interaction between signaling pathways and immune balance, and construct network model of “disease-syndrome-gene-signaling pathways”,and explore mechanism of the curative effect of traditional Chinese medicine in vitro and in vivo . We hope to provide new ideas for interpretation the mechanism of traditional Chinese medicine treatment.
