关节骨侵蚀是RA中晚期的关键病理特征,延缓甚至阻断骨侵蚀是RA 治疗的重要靶标之一。本课题在艾灸治疗RA研究取得重要进展的基础上,依据“骨代谢失衡是RA骨侵蚀的重要基础,OB/OC是骨代谢平衡的关键要素,DVL2交叉调控Wnt/NF-κB信号通路与骨代谢密切相关,艾灸治疗RA具有良好的抗炎和骨保护效应”等基本事实,提出“艾灸通过DVL2交叉调控Wnt/NF-kB信号通路,有效调节骨代谢平衡,促进RA骨重建”的研究假说,以中医针灸理论为指导,聚焦RA关键病理—骨侵蚀,围绕艾灸治疗RA目标效应—骨保护,以DVL2为研究切入点,以“细胞因子—Wnt/NF-κB信号通路—OB/OC骨代谢”为研究主线,运用慢病毒载体、基因芯片、免疫组化等方法技术,深入揭示艾灸治疗实验性RA骨保护作用的细胞因子调节、骨代谢平衡和信号通路调控机制,对于深化艾灸治疗RA的原理研究、丰富其科学内涵具有重要的价值和意义。

Bone erosion is a key pathological feature of RA in advanced, delaying or even blocking RA bone erosion is one of the important target in the treatment of RA. It is well known that the imbalance of bone metabolism is a major reason for bone erosion in RA,OB/OC is a key element of bone metabolic balance,DVL cross regulation of the Wnt and NF-kB signaling pathway,and moxibustion in the treatment of RA has good anti-inflammation and bone protective effect.This study is based on the important progress for the effect of moxibustion therapy on RA,put forward a hypothesis is that moxibustion can cross regulation of Wnt and NF-kB signaling pathway by DVL,effectively rebalance of bone metabolism and promote bony remodeling in RA.This study take TCM acupuncture theory as the guidance,focusing on the key pathological is bone erosion,according to the core effect of moxibustion treatment is bone protection in RA,with DVL2 as the point, the main line is “cytokine—Wnt/NF-kB pathway—bone metabolism(OB/OC)”,using lentiviral vector,gene chip,immunohistochemistry and other modern research methods.To reveal the regulation mechanism of moxibustion in the treatment of experimental RA for cytokines,bone metabolism and signaling pathways.For this study,the important value is to deepen and rich the scientific connotation of acupuncture and moxibustion treatment on RA.
