肿节风用药历史悠久,其药材及制剂已广泛应用于临床,研究已表明糖类聚合物是肿节风中的重要活性成分之一,但有关肿节风及制剂的质量控制仍以单一(类)小分子成分为主,糖类聚合物尚未被作为质控指标。本课题拟针对肿节风中的糖组,采用糖类结构解析的技术研究不同来源的肿节风糖组的关键结构特征,构建单糖、寡糖、多糖、糖复合物结构特征的多元指纹图谱,包括高效离子交换色谱、高效凝胶渗透色谱、核磁共振波谱、荧光辅助糖电泳等指纹图谱,并以抗肿瘤活性为肿节风糖组的活性评价指标,采用灰度关联度分析及多元线性回归等数理统计方法解读基于糖组的指纹图谱与抗肿瘤活性的谱效关系,并确认肿节风中活性糖类的特征标识,为提升肿节风及其制剂的质量标准提供科学依据。通过肿节风糖组的解码及谱效关系的研究可形成一条普适性的基于糖组化学特质的中药糖类组分的质控评价体系,为其他中药的整体质量控制提供参考及借鉴,对中药的市场监管具有重要的现实意义。

Sarcandra glabra has been used as medicine for a long history, its related pharmaceutical preparations have been widely used in clinic. Carbohydrates have been studied as one of the most important active components. But the quality control points in medicinal materials and products were mainly focus on single compounds. Carbohydrates haven’t been studied in the herbal’s quality control. The project will first aim at glycome decoding in Sarcandra glabra according to chemical, instrumental, and biological methods and technology. Multi-fingerprints based on the structural features from free, polymetric carbohydrates, glycans released from glycoconjugates will be constructed. The multi-fingerprints include high-ion exchange chromatography fingerprint, high performance gel permeation fingerprint, nuclear magnetic resonance spectroscopy fingerprint, fluorescence-assisted carbohydrate electrophoresis fingerprint, and so on. The activity of glycome will also be determined by antitumor evaluation. Then the spectrum-effect relationship will be analyzed based on the data from multi-fingerprints and activity determination results. Herbal carbohydrate markers will be finally obtained from spectrum-effect relationship analysis and activity verification. It will be an important scientific support for improving the quality control of Sarcandra glabra and related products. The results also provide a strategy for overall quality control of herbal medicine based on the structural features of carbohydrates. This work will have a positive impact and practical effect of supervising on traditional Chinese medicine.
