近五年,申请人围绕消化道疾病发生发展的表观遗传和信号通路机制开展了系列研究。取得如下成果:①揭示了胆汁酸诱导Barrett食管发生恶变的分子机制,为防治Barrett食管恶变提供潜在靶点。②发现了关键分子STAT3、COX2、TMEFF2和SHP-1在Hp诱导的胃炎恶变中扮演重要角色。③证明了miR-194作为大肠腺瘤切除后再发的预警靶点的价值。以上成果发表SCI论文29篇,其中第一或通讯作者19篇 (2013 IF合计101.9分,单篇最高13.3分;SCI论文他引167次,单篇最高46次)。获教育部“新世纪”人才计划支持和上海市的多个荣誉称号。作为负责人承担2项NSFC课题。以第四完成人获2014年上海市科技进步一等奖和2013年教育部科技进步一等奖。后续研究的重点是肠菌失调导致肠黏膜细胞自噬减少和DNA损伤而参与黏膜恶变的调控机制;有望发现相关的预警与早诊标志分子。

英文摘要

My research work mainly focused on the signaling pathways and epigenetic changes during the initiation and progression of digestive disease in the past five years. We have made the following progress. ①We showed that TGR5 (a G-protein-coupled receptor) may participate in bile acid taurodeoxycholic acid (TDCA)-induced the malignance of Barrect esophagus and the regulatory mechanism, providing potential prevention and treatment targets for the malignance of Barrect esophagus. ②We focused on the function of STAT3、COX2、TMEFF2 and SHP-1 in Hp (Helicobacter pylori) induced the malignance of gastritis. ③ We found that mir-194 may act as an effective biomarker to predict colorectal adenoma recurrence. Our research may provide the important theoretical basis of application from bench to bed in digestive disease. So far, I have published 29 SCI articles, among which I am the first/corresponding author in 19 articles. The 2013 IF(Impact Factor) amounts to 101.9, and the highest IF of single article is 13.3.My SCI articles are cited 167 times by others ,and the most quoted article is cited 46 times. At the present, I am responsible for one of the national natural science fund major research plan cultivation project and one of national natural science fund general project. My research work is supported by the Shanghai Rising-Star Program, The Program for Professor of Special Appointment (Eastern Scholar) at Shanghai Institutions of Higher Learning and the dawn planned project of Shanghai Municipal Education Commission, Education and Development Foundation. I am supported by the program for 2013 Ministry of Education selected the New Century Talents Support Plan, as a major member to be selected in the Program for Changjiang Scholars and Innovative Research Team in University in 2011 and was honored 2013 Ministry of Education selected advance of science and technology(First Prize)(Rank 4). I also was honored Shanghai Science and Technology Progress Award (First Prize)(Rank 4). Next, we will focus on the role of Fn (Fusobacterium nucleatum, F. nucleatum), which may decrease the Autophagy and increase DNA damage, in the malignance of colon tissues. We expect to find some molecules were potentially to be developed as effective diagnostic biomarkers for the malignance of digestive disease in our research work.
