胚胎与母体之间适宜的相互作用是妊娠维持和母胎健康的重要保障。复发流产和子痫前期等疾病发病率高,母儿危害大,而有效预测和干预手段非常匮乏。对其发病机制的研究是国际生殖健康领域的热点和难点。本项目基于“母-胎相互作用决定妊娠结局”这一核心理念,围绕胚胎植入、子宫基质-蜕膜转化、滋养层细胞分化、母-胎免疫耐受等关键生理环节展开生物医学-生命科学-化学的多学科交叉研究:1)拟揭示各个生理时期维持正常妊娠过程的关键调控分子及通路,2)阐释上述生理事件调控紊乱导致复发流产和子痫前期等妊娠疾病的分子基础。3)建设基于中国人群的规范化妊娠疾病临床资源库,4)挖掘复发流产和子痫前期的新型遗传及表观遗传标记,用于疾病的临床诊断及亚型分类。通过本项目的实施,力争阐明母-胎相互作用的分子机制及其在重要妊娠疾病发生发展中的作用,在国际竞争中占领领域的制高点,为探索妊娠疾病早期诊断和干预的综合策略提供新的视角。

英文摘要

The well-regulated maternal-fetal interaction is essential for normal pregnancy and fetal growth. Recurrent spontaneous abortion (RSA) and preeclampsia (PE) are major pregnancy complications with serious clincal and social impact. The efficient prediction and prevention approaches for these diseases are largely lacking. It has been well believed that these gestational diseases are rooted in the abnormal maternal-embryo interaction since early pregnancy, whereas the molecular basis remains unclear and has now become the central concerns in the field of reproductive health. The present research project will combine bio-medical, chemical and genetic approaches to dissect the molecular basis that govern major pregnancy events including embryo implantation, uterine decidualization, trophoblast differentiation, and pregnancy immune tolerance, and to provide molecular clues in explaining how the abnormalities in these events contribute to RSA and PE. We also aims to establish the most comprehensive, standardized clinical sample library for pregnancy diseases in Chinese population, and to use the sample library to further discover novel biomarkers (at DNA, RNA and protein levels) for diagnosing and predicting these diseases. Though this collaborative 5-year project, we aim to provide deeper molecular understandings as well as novel diagnostic and preventive approaches for RSA and PE.
