肺癌转移与肿瘤微环境密切相关,TAMs-PI3K是调节肿瘤微环境的关键环节。中药复方抑肺饮是课题申请人防治肺癌的经验方,前期研究发现抑肺饮具有抑制肺癌细胞增殖,抗肺癌复发转移等作用,其调控机制与多条炎性信号通路、细胞因子及肿瘤微环境相关。本课题将针对TAMs-PI3K环节,研究抑肺饮早期干预肿瘤微环境、抑制肺癌转移的作用机制。本研究将运用荧光素酶标记的Lewis肺癌细胞建立肺癌原发、转移模型,通过转录组学、免疫组化、流式细胞分析、荧光定量PCR等技术发现抑肺饮调控TAMs-PI3K环节的关键位点,并从肿瘤微环境慢性非可控性炎症、免疫抑制及Cross-talk三个方面重点揭示抑肺饮通过TAMs-PI3K环节早期干预肿瘤微环境、抑制肺癌转移的作用机制,从而为抑肺饮的临床应用提供实验依据,为揭示中药复方抗肿瘤转移的作用靶点及其机制提供新的途径与思路。

The metastasis of lung cancer is closely related to tumor microenvironment. TAMs-PI3K plays a key role in regulating tumor microenvironment. Our previous study showed that Chinese medicine formula “YiFei Yin” can inhibit proliferation and metastasis of lung cancer by regulating several inflammatory signal pathway, cytokines and tumor microenvironment. In this research, we focus on the mechanism of “YiFei Yin” inhibiting metastasis of lung cancer by early intervention in tumor microenvironment based on TAMs-PI3K. We will discuss the mechanism from three aspects: chronic uncontrolled inflammation, immunologic inhibition and cross-talk in tumor microenvironment by using mouse model of primary and metastatic lung cancer and molecular biology technology (transcriptome, immunohistochemistry, flow cytometry and real-time PCR, etc). Our proposal is innovative and significant, because it will not only provide an experimental basis for clinical application of “YiFei Yin” but also help in developing new strategies for unraveling targets and the mechanism of resisting metastasis of lung caner by Chinese medicine formula.
