病毒与介体互作研究是植物病毒研究热点之一。番茄褪绿病毒(Tomato Chlorosis Virus, ToCV)是我国番茄生产的重要威胁且没有抗病品种,建立ToCV新型绿色防控技术已经刻不容缓。本研究以ToCV CPm基因差异调控不同生物型烟粉虱传播病毒效率为基础,通过构建不同CPm突变体和异源CPm的侵染性克隆,以本氏烟为材料验证相关侵染性克隆和不同生物型烟粉虱传播ToCV的效率差异,结合荧光标记证实ToCV病毒粒子在烟粉虱体内滞留位点和时间差异,通过获毒前后滞留位点转录组测序明确传毒烟粉虱自身调节相关基因,并以相关基因为探针,在滞留位点基因文库中对靶标基因进行筛选和鉴定,进而结合酵母双杂交系统验证不同CPm结构域与靶标基因的互作,确定互作的关键位点和有效靶标基因,并利用RNAi技术对有效靶标基因进行生物学功能解析,为ToCV新型绿色防控、高效阻断病毒传播药物设计和抗病育种基础。

Understanding of interaction of virus with its transmitting vector, would be help to develop novel preventing technology. Currently, it is emergency to prevent tomato chlorosis virus (ToCV) for threating the crops of Soanaceae. Our preliminary studies show the capacity of transmission of ToCV by Q and B biotype Bemisia tabaci is different probably because of differently mediated by ToCV CPm. This project intends to construct infectious clones of ToCV CPm mutant and ToCV mutant with heterogenous CPm, the transmission capacity by B and Q biotype Bemisia tabaci, combine with the site and temporal differences of virions of ToCV mutants in vector by fluorescence labeling, screening and validating the proteins of vector interacting with CPm by transcriptome, yeast two-hybrid and RNAi. The scientific significance of this project will help to resistant breeding, developing novel chemicals for blocking viral transmission.
