外泌体miRNA的发现成为血管性痴呆(Vascular Dementia,VD)早期诊断与治疗靶点研究热点,为中药复方多靶点干预机制研究提供了新思路。既往研究显示参麻益智方治疗轻中度VD有良好效果,在此基础上,课题组进一步提出“对VD外泌体 miRNA的特异表达及其miRNA-mRNA网络的有效调控是参麻益智方防治VD的主要作用靶点之一”的假说。因此,本课题利用外泌体 miRNA芯片技术、生物信息学方法,筛选VD目标miRNA,构建miRNA-mRNA网络,观察参麻益智方对该miRNA及miRNA-mRNA网络的调控作用。同时通过该研究,为与VD早期诊断相关的分子生物标记物的发现提供依据。
英文摘要
The discovery of exosome miRNA has become a hot topic in the early diagnosis and therapeutic targets of vascular dementia. It provides a new idea for the research in multi-target intervention mechanism of Traditional Chinese Medicine compound prescriptions. Previous studies have shown that Shenma Yizhi prescription has a good effect on the treatment of mild and moderate VD. Based on this, research team propose the hypothesis "The effective regulation of the specific expression of exosome miRNA and miRNA-mRNA network in VD is one of the main target of Shenma Yizhi prescription on the prevention and treatment of VD. " Accordingly,this subject use exosome miRNA chip technology、biological information method to screen VD specific miRNA and construct miRNA-mRNA network. We observe the regulatory effect of Shenma Yizhi prescription on the miRNA and miRNA-mRNA network. At the same time, we provide the basis for the discovery of molecular biomarkers associated with the early diagnosis of VD through the study.
