植物类受体激酶是细胞表面介导信号转导的关键组分,在调节植物生长发育及环境响应等方面发挥关键作用。拟南芥ERECTA家族类受体激酶识别EPF多肽信号分子,调控植物器官生长及气孔发育等重要生物学过程。但是,ERECTA家族受体在识别EPF信号分子并被激活后,如何将信号传导到胞内并调控植物生长发育目前还很不清楚。申请人拟通过磷酸化蛋白质组学分析、互作蛋白分离以及遗传互补筛选等方法系统鉴定ERECTA受体的下游信号组分,进而结合遗传、生化等手段研究所分离的信号组分在ERECTA途径中的功能及作用机制,对于全面揭示ERECTA信号途径、了解植物类受体激酶的功能及信号转导机制、认识植物生长及气孔发育的调节机制都具有重要科学意义。申请人近几年一直在对ERECTA信号途径进行相关研究,已经取得了较为显著的研究成果,同时积累了研究材料,建立了研究体系,为开展本申请中的工作奠定了坚实基础。
英文摘要
Plant receptor-like kinases (RLKs), the key signal transduction components on the cell surface, play important roles in regulating plant growth, development, and response to the environment. The Arabidopsis ERECTA family RLKs play pivotal roles in regulating plant organ growth and stomatal development by recognizing the EPF family of signaling peptides. But how ERECTA receptors transduce EPF signals to intracellular components is largely unknown. In this proposal, we will use phosphoproteomic approaches, protein-protein interaction screening, and genetic complementation screening to systematically identify the signaling components downstream of ERECTA receptors. Furthermore, genetic and biochemical approaches will be used to elucidate the functions of the identified components in the ERECTA signaling pathway. This study will significantly advance our understanding of the ERECTA pathway and the signal transduction mechanisms of plant RLKs. The applicant has productive publications in this area during the past several years, and as a result accumulated a large number of useful materials and constructed many effective research systems, which will significantly facilitate the future work.
