毛竹开花的不确定性威胁竹产业安全和生态安全,因此研究毛竹开花调控机理具有重要意义。尽管人们在毛竹开花生理机制研究中取得一些进展,但其分子作用机制仍不清楚。本课题组对毛竹用RNA-seq分析发现11个Dof转录因子参与了毛竹的开花调控通路,随后的免疫印记和原位杂交表明PheDof1呈现花器官和开花发育阶段特异表达,暗示Dof参与了毛竹开花调控。在此基础上,以Dofs基因特别是PheDof1为研究对象,研究这些基因在竹子开花启动与进程中的表达规律,探索敲除或过表达Dof对下游各个途径成员表达的影响,用RNA-seq、CHIP-seq、Y1H和EMSA搜寻并鉴定调控的开花下游靶基因,查验干旱胁迫处理对Dof以及下游基因(包括直接结合的和间接调控的)表达的影响, 最终揭示Dof调控开花的分子机制。这对揭示多年生植物开花机理有重要的科学意义,对毛竹的杂交育种和分子育种具有一定的理论指导意义。

英文摘要

Moso bamboo (Phyllostachys edulis) is one of the most important economic bamboo species, but the unpredictable flowering feature has threatened the bamboo growth and development. Although some researches have been reported in the physiological mechanism on moso bamboo flowering, the molecular mechanisms still remain poorly understood. Our previous research of the RNA-seq and expression profile analysis has showed that 11 Dof transcription factors might be involved in regulatory pathways of moso bamboo flowering. The results of western blotting and in situ hybridization indicated that PheDof1 major expressed in particular flower organs and development stages. Based on these previous results, in this project, 11 Dof genes, especially the PheDof1 gene, were chosen to study the expression and co-expression patterns of Dof genes and other downstream flowering regulation genes at flower initial and developmental stages by quantitative reverse transcription PCR assay. Further, the downstream target genes of PheDof1 gene will be screened by ChIP sequencing using the knockout and over-expression Dof1 gene and will be identified by EMSA and yeast one hybrid assay. In addition, drought stress treatment in transgenic plants will be included to check the expression of Dofs and downstream target genes. It will be expected to reveal the downstream regulatory network and elucidate the molecular mechanism of Dof transcription factors affecting on the downstream targeted genes. It will possess vital scientific significance for the study on the flowering mechanism of perennial plants.
