中药因其多靶点调节优势在治疗性血管新生领域引人注目。补脏通络方立意补脏通络、祛瘀生新,治疗糖尿病血管病变作用良好,前期研究表明,该方以eNOS为靶点对糖尿病血管病变具有潜在的治疗性血管新生作用,但因缺乏在体实验研究,其作用效果及深入机制不明。研究表明,糖尿病状态下,VEGF/Akt/eNOS信号通路活化受损是导致骨髓EPCs动员障碍及血管新生障碍的重要机制。基于以上思路,本课题拟采用糖尿病下肢缺血动物模型为研究对象,观察补脏通络方改善糖尿病缺血组织的治疗性血管新生作用,明确该方对糖尿病VEGF/Akt/eNOS信号通路及骨髓EPCs动员的影响,初步探讨其治疗性血管新生作用机制与其干预糖尿病VEGF/Akt/eNOS信号通路功能及骨髓EPCs动员障碍的关系,为进一步阐明补脏通络方治疗糖尿病血管病变的机制及探讨中医补脏通络、祛瘀生新法与治疗性血管新生的关系提供依据。

Chinese Medicine is remarkable in the therapeutic angiogenesis because of its multi-targeted regulation. BuZangTongluo Decoction has good effect on diabetic vascular disease in the conception of BuZangTongluo and QuYuShengXin. Previous studies showed that BuZangTongluo Decoction has the potential therapeutic angiogenesis with eNOS as a target, but due to lack of the study in vivo , it’s effect and deep mechanism is unknown. Researches showed that the activation of VEGF/Akt/eNOS pathway is an important mechanism in leading to the disorder of bone marrow-derived endothelial progenitor cells mobilization and diabetic angiogenesis in the diabetic state. Based on the above ideas, this topic proposed the diabetic lower limb ischemia animal models as the research objects, to observe the therapeutic angiogenesis of BuZangTongluo Decoction on diabetic ischemic tissues and explicit the effect of BuZangTongluo Decoction on diabetic VEGF/Akt/eNOS signal pathway and bone marrow-derived endothelial progenitor cells mobilization, for further clarifying the mechanism of BuZangTongluo Decoction on treating diabetic vascular disease and exploring the relationship between the Chinese Medicine theory of BuZangTongluo and QuYuShengXin with the therapeutic angiogenesis.
