细胞正常和病理行为预测及干预是系统生物学研究的核心任务之一，同时也是从系统水平研究中医药理论和作用机制的前沿领域。然而目前在开展基于真实环境和系统水平的细胞动力学模拟方面，尚缺乏基础理论和相应算法。本项目拟在一个整合的框架下，采用统计力学、流体力学、分子动力学以及系统控制等理论，以正常和病理细胞的发生、发展和治疗过程为研究对象,构建精确的多细胞微环境和细胞内、细胞间分子网络动力学系统;同时将该理论体系应用于典型的多尺度疾病发生模型-心肌肥大病,及其在中药(丹参、黄芪、芍药等/单体成分等)治疗下的药物分子传递、吸收、网络靶标结合以及细胞命运演化的系统动力学研究;旨在最终开发出基于复杂系统理论的细胞行为预测方法和数学模型。本系统的成功开发将为从系统水平阐明中药和机体的相互作用、中医药基础理论及中医药治疗新策略研究提供理论基础。

The prediction and interference of cell behaviors is a big challege for systems biolgoy. The sucessful prediction of cell behaviors will provide a significant support for therapeutic treatments of disease cells. However，the cellualr simulation and modeling based on the intergrative framework is scarces presently. The present application in devoted to develop an intergrative model using statistical mechanics, fluid mechanics and molecular dynmics, to investigate the normal and diseased cell growth,development as well as the treatment by drugs. Meanwhile, the drug multiscale tansfering, absorption and receptor binding are also studied and finally to provide important therapeutic methods for treatment of diseases. The developed models will also be applied to the cardiac hypertrophy system in oder to clarify the cell contact,environment and therapeutical treatments by Traditional herbal medicines. The successful results of this work will provide important theory and tools for integrative biology as well as the undertannding of Traditional Chinese Medicine.

细胞内分子网络相互作用预测及动力学模拟是系统药理学研究的关键点之一，同时也是从系统水平研究中医药理论和作用机制的前沿领域。然而目前在开展基于系统水平的中药分子机制研究方面，尚缺乏基础理论和相应算法。本项目在一个整合的框架下，采用统计物理、分子动力学以及系统控制等理论，以正常和病理状态下细胞分子网络为研究对象，构建了药物直接靶点预测方法WES、药物基元动力学模拟方法ESDA等，并建立了中药系统药理学分析软件TCMSP。同时将这些理论和方法应用于多种复杂疾病（如心脑血管疾病，癌症等）和中药治疗分子机制的模拟分析研究中。本项目的完成为从系统水平阐明中药和机体的相互作用、中医药基础理论及中医药治疗新策略研究提供了新思路和新方法。
