血管周脂肪分泌adiponectin发挥抗血管收缩功能。巨噬细胞聚集可导致是脂肪细胞adiponectin分泌下降。前期结果证实加味二至丸可改善卵巢切除大鼠胸主动脉管周脂肪抗血管收缩功能并抑制管周脂肪炎症细胞浸润。本研究在整体、器官、细胞分子水平进一步探索加味二至丸改善更年期大鼠胸主动脉管周脂肪抗血管收缩功能的分子机制。首先,在更年期大鼠模型中应用qPCR、免疫荧光、流式细胞术等技术,观察加味二至丸对更年期大鼠胸主动脉管周脂肪抗血管收缩功能、巨噬细胞的聚集与极化、adiponectin分泌的影响;其次,在细胞模型中应用细胞共培养、流式细胞术、Tranwell小室、siRNA等方法观察加味二至丸含药血清对脂肪细胞adiponectin分泌、巨噬细胞聚集及M1型极化的影响及不同雌激素受体在其中的作用。本研究将为绝经后女性心脑血管疾病的中医药防治提供新思路。

Perivascular adipose has anti-contractile function by secreting adiponectin. Macrophage accmulation leads to less adiponectin expression in adipose tissue. Our previous studies showed erzhi pills combined with epimedium could improve the anticontractile function of periaortic adipose tissue in ovariectomized rats and inhibit inflammatory cell accumulation. This study investigates its molecular mechanisms. By qPCR, we observe the effects of erzhi pills combined with epimedium on the secretion of adiponectin in periaortic adipose tissue. By immunofluorescence and flow cytometry we also investigate the effects of erzhi pills combined with epimedium on macrophage accumulation and M1 polarization in periaortic adipose tissue in menopausal rat models. Then we study the inhibitory effects of serum containing erzhi pills combined with epimedium on the secretion of adiponectin in adipocytes and on migration of macrophages by cell migration assays and transwell experiments. The involvement of estrogen receptors in the inhibitory effects on migration of macrophages is also investigated by siRNA and transwell assay. The involvement of estrogen receptors in M1 macrophage polarization is also investigated by flow cytometry assay and siRNA. This study provides novel ideas for treating menopause associated cardiovascular diseases.
